| Taraxacum kok-saghyz Rodin(TKS)has been studied as an alternative source of natural rubber because its roots are rich in high-quality rubber.In addition to being rich in natural rubber(NR),TKS also contains inulin and its leaves contain antioxidant ingredients such as chicoric acids and chlorogenic acids.It is the first time to explore the possible of TKS for potential applications to improve the value of TKS.In this study,UHPLC-Q-Orbitrap HRMS was used to conduct non-target screening and identification of the unknow components in TKS,and preliminary explored the mass spectrometry fragmentation law of some compounds under the APCI ion source.The optimized extraction conditions of target components(pentacyclic triterpenoids and sterols)obtained from non-targeted screening by response surface methodology.An ultrasonic/microwave-assisted extraction-ultra-high performance liquid chromatography-tandem mass spectrometry method for the determination of target components in TKS was developed,and the difference in content of target components between TKS and common dandelion was also compared.According to the screening and identification,164 chemical compounds were obtained,mainly including terpenoids,coumarins,flavonoids and organic acids.The optimized ultrasonic/microwave-assisted extraction conditions with liquid-to-material ratio of 45 mL/g,microwave power of 420 W and ultrasonic power of 560 W were obtained.The detection limit and quantification limit of the target components of the UHPLC-MS/MS method are 0.5-5.0 ng/g and 1.0-10.0 ng/g,respectively,and the intra-day and inter-day accuracy are less than 5.5%,which can be applied to TKS target components detection.The content of the target components(9.44 mg/g)in TKS was significantly higher than that of the common dandelion(3.35 mg/g),and the relative content of β-amyrin was the highest in TKS.There are many kinds of medicinal components of TKS,and the content of pentacyclic triterpenes and sterols is relatively high,which has high potential medicinal value. |
