| Objective:Data mining is carried out for the urticaria cases diagnosed by Professor Xu Aiqin in the dermatology clinic.The main platform for data mining is the ancient and modern medical record cloud platform(2.3.5),and the ultimate purpose is to count,analyze and summarize Xu’s experience in treating urticaria and drug filtering.,and excavated the core drugs of Xu Shi’s treatment of urticaria based on clinical medical records.Conduct network pharmacology research on the core drug pairs displayed in the data mining,and search and discuss the main traditional Chinese medicine components,targets and pathways of the core drug pairs in the treatment of urticaria.Methods: 1.Data mining: Collect the medical records of urticaria diagnosed by Professor Xu Aiqin of the Department of Dermatology of Wuhan Hospital of Traditional Chinese Medicine in 2021,sort out and screen all the collected medical records in a standardized way,and finally complete 174 medical records that meet the requirements,and then import the screened medical record data into the ancient and modern medical record cloud platform(2.3.5)for relevant information analysis.The main contents include: the frequency of general data,the frequency of TCM syndromes,the frequency of TCM treatment methods,the frequency of drugs,association rules,etc.,as well as the mining of core drugs,and finally the interpretation and elaboration of TCM theory.2.Network Pharmacology: To study the drugs that have been excavated in the previous step,the main steps are:(1)Find the active ingredients and corresponding targets of core drug pairs with OB>30% and DL>0.18 in the TCMSP chemical professional database.(2)Use the four disease databases of Gene Cards,Drugbank,TTD,and OMIM to search for disease targets,find out the common targets of the disease and the core drug pair,and construct the intersection map of the core drug pair and the common target of the disease.(3)Use Cytoscape software to construct and draw relevant network diagrams,and find out the traditional Chinese medicine ingredients that play the most important role in the treatment of urticaria.(4)The STRING platform was used to construct the PPI network,and then imported into Cytoscape for topology analysis to find the key targets of the core drugs for the treatment of urticaria.(5)Use the Bioconducter software package to run the analysis in the R language to obtain the GO and KEGG analysis results.(6)Input disease-drug intersection targets in Metascape platform,and output MF,CC,BP and KEGG analysis results in GO.Results:1.Data mining results:(1)The results of frequency analysis of general data show that there are 112 females,accounting for 64.37%;62males,accounting for 35.63%;the majority of the population is 30-39 years old.(2)The results of frequency analysis of TCM syndromes showed that there were 12 TCM syndromes in total,and the top three syndromes were the syndrome of disharmony between ying and wei,the syndrome of yin deficiency and blood dryness,and the syndrome of spleen deficiency and fire exuberance.(3)Frequency analysis of traditional Chinese medicine treatment methods,the results show that there are 14 kinds of treatment methods,among which the main ones are reconciling yingwei,expelling wind and relieving itching,nourishing yin and clearing heat.(4)The results of drug frequency analysis showed that there were 156 traditional Chinese medicines used,and the top 10 ones were Fangfeng,Nepeta,Poria,Arctium,Coix Seed,Yam,Atractylodes,Citrus aurantium,Licorice,and Jujube.The five flavors are mostly sweet and acrid;they are mostly attributed to the spleen and lung meridians;the functions of traditional Chinese medicine are mainly to clear away heat and detoxify,disperse wind-heat,and penetrate rash.(5)The results of correlation analysis showed that the overall drug use was centered on Fangfeng,the highest support was in the Nepeta,Fangfeng group,and the highest confidence was in the Nepeta,Fangfeng,Chinese yam,and Fangfeng group.(6)The results of cluster analysis showed that the eight groups of commonly used Chinese herbs for the treatment of urticaria were:(1)Forsythia;(2)Bran fried Atractylodes Rhizoma,Bran fried Citrus aurantium;(3)Malt,Monascus;(4)Schisandra,Rehmannia glutinosa,Eclipta Zhenzi;(5)Dandelion,Poria,Licorice;(6)Codonopsis,tangerine peel,honeysuckle;(7)Angelica,Ophiopogon japonicus,Taizishen,Astragalus,licorice,cinnamon,white peony;,Jujube,Atractylodes,Citrus aurantium.(7)Complex network analysis and core drug mining: After data mining and analysis,the final core traditional Chinese medicines are: Fangfeng and Nepeta.2.Network pharmacology results:(1)28 active ingredients of core drug pairs and 202 corresponding targets were obtained through the TCMSP database search,and imported into Cytoscape to construct a network of "Fangfeng-Nepeta-active ingredients-target gene".(2)Searching "urticaria" in four related disease data such as Gene Cards and TTD,a total of 994 disease-related targets were obtained,and a total of 82 common targets were obtained after intersecting with drugs,which were imported into Cytoscape to generate "drug-Active ingredient-disease-target gene" network diagram,according to Degree,the key ingredients of core drugs for the treatment of urticaria include quercetin,luteolin,wogon scutellariae,5-O-methylviaminol,etc.(3)After creating the PPI network,key targets were obtained: IL6,AKT1,TNF,IL1 B,TP53,MMP9,CASP3,PTGS2,etc.(4)GO enrichment analysis was carried out,in which GO-BP analysis showed that it was mainly involved in inflammatory response,cell response to nitrogen compounds,positive regulation of protein phosphorylation,etc.;GO-CC analysis showed that potential targets were mainly located in membrane rafts,The outer membrane of the organelle,the outer side of the plasma membrane,and the perinuclear region of the cytoplasm;GO-MF analysis results showed that the molecular functions were mainly related to cytokines,G proteins and adrenergic receptors.(5)KEGG enrichment analysis was performed to obtain key signaling pathways such as PI3K-Akt,NF-k B,FCεRI,Th17,and AGE-RAGE.Conclusion:1.Based on clinical outpatient medical records,Master Xu’s drugs,drug combinations,and core drugs for the treatment of urticaria mainly focus on reconciling Yingwei and dispersing external pathogens,and they are closely related to the lungs and spleen in terms of meridians.The composition of the core drug pair is: parsnip,nepeta.2.Parsnip,Nepeta,etc.may use a variety of complex mechanisms to reduce the inflammatory response and oxidative stress response to skin lesions,and jointly mobilize the body’s immune system,which in turn has a healing effect on urticaria. |
