Analysis Of Professor Yang Baijing’s Medication Patterns And Mechanisms In The Treatment Of CG Based On Data Mining And Network Pharmacology

Object: With the help of the traditional Chinese medicine inheritance assistant platform(V2.5),to explore the clinical medication characteristics and prescription rules of essor Yang Baijing,a famous Chinese doctor in the Corps,in the treatment of chronic gastritis,and to sort out and summarize its core drugs for the treatment of chronic gastritis.It also uses network pharmacology to analyze the core drugs,and expounds the characteristics of Professor Yang Baijing’s treatment of chronic gastritis from a microscopic perspective,so as to provide reference for the clinical treatment of this disease in traditional Chinese medicine.Methods: In the HIS system of an affiliated hospital of Shihezi University School of Medicine,clinical cases of chronic gastritis treated by Professor Yang Baijing in outpatient clinics from June 2019 to October 2021 were collected,and the cases were screened and standardized,then entered into the TCM Inheritance Assist System(V2.5)platform for clinical data analysis and mining of Professor Yang Baijing’s drug use patterns.Then,from the commonly used herbal medicines mined,key herbal medicines similar to monarch and minister were selected under the guidance of Professor Yang Baijing,and the active ingredients of the herbal medicines and corresponding target genes and chronic gastritis disease target genes were retrieved in the corresponding databases;the retrieved drug target genes and disease target genes were processed in Venny software to obtain the intersection genes of both,and then the STRING database and Then,we used STRING database and Cytoscape software to analyze the intersecting genes to obtain the key active ingredients and key genes;we used Metascape database to analyze the intersecting genes by GO and KEGG enrichment to obtain the relationship between drug action and disease targets;finally,we selected the top key active ingredients and key genes for molecular docking verification.Result:1.Professor Yang Baijing is treating the "disharmony between the liver and stomach,spleen and stomach weakness,spleen and stomach damp-heat syndrome,stomach yin deficiency syndrome,and stomach collateral stasis syndrome",and the distribution of the diagnosis and treatment of these five TCM syndrome types.2.From high to low,the frequency of the use of traditional Chinese medicine slices ranked in the top13 Chinese medicine flavors: poria cocos,ginger pinellia,scutellaria baicalensis,bupleurum chinense,betel nut,radix pseudostellariae,rhubarb,evodia rutaecarpa,neem,radish seed,large belly bark,atractylodes macrocephala.Because licorice is used as a "medicine" in the formula-a mixture of various drugs,and its frequency is very high,licorice is excluded from the core drugs.3.According to the analysis of formula rules(rule analysis),there are 6 pairs of commonly used drug pairs,including Pinellia ternate-Poria cocos,Radix Pseudostellariae-Poria cocos,Fructus Evodiae officinalis-Poria cocos,neem-Poria cocos,betel nut-Poria cocos,and neem-betel nut.4.The core drug formulations were obtained through analysis: Radix Pseudostellariae,Radix et Rhizoma Rhei,Melia azedarach,Rhizoma Pinelliae,Areca catechu,Radix Scutellariae baicalensis,Cortex Peribelli,Poria cocos,Radix Bupleuri,Fructus Evodiae,Semen Raphani,Atractylodes macrocephala.5.According to the new formula analysis,three new formula combinations were obtained.Square 1:Schisandra chinensis,Dabelli peel,wine rhubarb,mountain ophiopogon japonicus;Formula 2: gardenia,salvia miltiorrhiza,wine scutellaria baicalensis,mint,prepared rehmannia;Fang 3: Radix Rehmanniae,Semen Raphani,Cortex Moutan,Fructus Evodiae,Radix Bupleuri,Cortex Dalbergiae.6.Network pharmacological analysis: select the monarch and subordinate drugs in the core drug group,extract the target genes of drugs and diseases,obtain 22 intersection genes,and screen 14 key genes;According to GO and KEGG analysis,the drug mainly acts on inflammation and immune regulatioregulating cancer pathway,inflammatory bowel disease pathway,AGE-RAGE signal pathway,leishmaniasis,IL-17 signal pathway and other signal pathways.Conclusion:1.Professor Yang Baijing believes that the pathogenesis of chronic gastritis is "due to the abnormal function of the spleen and stomach lift,the essence of water and grain can not transform into qi,blood and body fluid,resulting in the deficiency of liver and spleen,resulting in qi stagnation,phlegm dampness,blood stasis and other symptoms,and stomach pain,fullness and other symptoms".2.Professor Yang Baijing’s treatment of chronic gastritis is based on the physiological characteristics of "spleen ascending and stomach descending,spleen transporting and stomach absorbing",focuses on the main symptoms,and takes invigorating the spleen and regulating qi,opening the pain and lowering the pain as the most basic treatment principles of chronic gastritis.3.In terms of drug selection,Professor Yang Baijing is good at using classic prescriptions.Through verification and investigation,he often uses traditional Chinese medicine for the treatment of this disease,such as invigorating the spleen and replenishing qi,drying and dampness,invigorating the spleen,soothing the liver and regulating qi,dissipating food and reducing turbidity,reducing adverse reactions and stomach.4.The core prescription summarized is mainly to treat chronic gastritis with "disharmony between liver and stomach" and "spleen and stomach weakness syndrome" as the entry point.Patients with chronic gastritis often have epigastric pain,and the qi of the central focus is not smooth.They often use Chaihu Shugan Powder to relieve liver and regulate qi and stomach;If the patient likes to warm but does not like to press,and is mentally tired,and if the spleen and stomach are weak,they should use Xiangsha Liujunzi Decoction and Huangqi Jianzhong Decoction to replenish qi and warm the spleen;If the stomachache is burning and painful,it seems that you are hungry and do not eat;The patient’s stomachache is full and pain has a fixed place,which cannot be cured over time.Most of them are Shixiao powder and Danshen drink to promote blood circulation and remove blood stasis.5.Through the analysis of the monarch and subordinate drugs in the basic prescription by network pharmacology,we can get the therapeutic mechanism of Professor Yang Baijing’s treatment of CG,which mainly acts on the key targets such as TP53,IL6,IL1 B,CXCL8,and regulates the signal pathways such as cancer and inflammatory bowel disease.
