Intestinal Microflora Analysis Of Damp-heat Polycystic Ovary Syndrome

Objective: To analyze the structural characteristics of intestinal microflora in patients with damp-heat polycystic ovary syndrome(PCOS)based on 16 S r DNA high-throughput sequencing technology.The intestinal microflora of the damp-heat PCOS group,the non-damp-heat PCOS group and the normal group were analyzed to explore whether there were differences in the intestinal microflora of PCOS patients and the normal group,and whether there were differences in the intestinal microflora of the damp-heat PCOS group and the non-damp-heat PCOS group,and analyze the differences,which could find a breakthrough point for the treatment of PCOS in traditional Chinese Medicine.Methods: Thirty cases of damp-heat type PCOS patients(A-PCOS,n = 10),non-damp-heat type PCOS patients(B-PCOS,n = 10),and healthy women(C,n =10)were collected,the general information,clinical biochemical index of the participants,the doctor of traditional Chinese medicine syndrome and other data of the three groups of patients were collected,fasting stool samples were collected from the patients in the morning,using high-throughput sequencing technologies,the bacterial 16 S r DNA V3-V4 fragments amplification,quantitative analysis,quantitative analysis of the characteristics of three groups of patients with intestinal microflora,intestinal microflora in patients with PCOS and healthy women whether there is a difference,and compare the damp-heat type PCOS to the non-damp-heat type PCOS intestinal microflora and differences,analyzed the composition,species and quantity of intestinal microflora in each group.Results :(1)In this study,30 subjects(damp-heat PCOS group,non-damp-heat PCOS group,and normal group)were included.There was no statistical significance in age,BMI of the three groups(P>0.05).AMH and AFC of PCOS group were significantly different from those of normal group(P<0.001).Compared with non-damp heat PCOS group,there was significant difference in the syndrome integral after damp-heat PCOS group.(P<0.05).(2)The α diversity analysis of the three groups showed no significant difference in Chao1 index,ACE index,Simpson index,and Shannon index.The Shannon index of the PCOS group was lower than that of the normal group,and the difference was statistically significant(P<0.05).However,there was no significant difference in αdiversity between the damp-heat PCOS patients and non-damp-heat PCOS patients(P>0.05).The results showed that the bacterial diversity of PCOS patients decreased.(3)The β diversity analysis of the three groups was carried out,and the differences among the three groups were compared by the PCOA method based on the weightde_Unifrac distance.Compared with the normal group,the richness and evenness of the PCOS patients showed statistically significant differences(P<0.05).In addition,the PCOA method based on Unweightde_Unifrac distance was used to compare the flora distribution among the three groups.Significant differences were found in the composition of intestinal flora between the damp-heat type PCOS and the non-damp-heat type PCOS and the normal group,that is,the richness of damp-heat type PCOS changed.(4)The number of Bacteroidetes,Bacteroidia,Bacteroidales,Bacteroidaceae in the PCOS group was significantly lower than that in the normal group(P<0.05).(5)LEf Se analysis of the three groups of bacteria showed that there were significant differences between the PCOS group and the normal group,and the abundance of Bacteroidetes,Bacteroidetes,Bacteroidetes,Bacteroidetes,Amycobacteria,Bacteroidetes in the PCOS group was significantly reduced.However,the abundance of Actinomycetes,Actinomycetes,Myridae and Myridae increased significantly.Megamonas,Unclassified,Rhizobiaceae,Ensifer,Luteimonas,α-Proteobacteria,Acidobacteria-GP6,Olsenella,Rhizobacteriaceae,Chromatiales,Nocardioides,and Novosphingobium in the damp-heat PCOS group increased significantly compared with the non-damp-heat group,and the composition of intestinal microflora was disorganized.Conclusions: 1.The intestinal microflora of PCOS patients was significantly different from that of healthy women(P<0.05);2.Compared with non-damp-heat PCOS patients,there are differences in the richness of bacteria in patients with damp-heat PCOS,and the composition of intestinal microflora in patients with damp-heat PCOS is significantly disturbed compared with that in the non-damp-heat group.
