| Background and purposeEndometriosis is one of the common causes of infertility in women of childbearing age,which brings a certain degree of trouble to women of childbearing age.Although assisted reproduction technologies brings hope to patients with EMs infertility,the effect of ART treatment in patients with EMs is still poor.In recent years,it has been found that the pathogenesis of infertility in EMs may be related to oxidative stress factors,but the relationship between them is still controversial.Oxidative stress means that when the body is subjected to harmful stimulation,the body tends to oxidize,which leads to the corresponding increase of protease secreted by neutrophils,which causes oxidative damage to proteins,lipids and deoxyribo nucleic acid in the body and produces a large number of oxidative intermediate products,resulting in functional damage such as cells and tissues.Follicular fluid is the microenvironment for the growth and development of oocytes,and the changes of its components in the environment are closely related to the quality of oocytes.In this study,the expression levels of oxidative stress markers superoxide dismutase and protein oxidation product advanced oxidized protein in follicular fluid of patients with EMs were measured to explore whether there is oxidative stress in follicular fluid of patients with EMs and the correlation between the expression levels of oxidative stress markers SOD and AOPP and the treatment of ART.MethodsThe follicular fluid of patients with EMs and infertility due to fallopian tube or male factors were collected from patients with EMs and infertility due to fallopian tube or male factors treated by ART in the first affiliated Hospital of Harbin Medical University from July 2019 to December 2019.According to the specified inclusion and exclusion criteria,the follicular fluid of 27 patients with EMs were collected as EMs group,and 31 patients with tubal or male factor infertility as control group.The patients included in the study above recorded their age,body mass index,years of infertility,serum basic follicle stimulating hormone,basic luteinizing hormone,basic estradiol,egg acquisition rate,MII egg rate,fertilization rate,cleavage rate,excellent embryo rate and so on.The contents of oxidative stress markers SOD and AOPP in follicular fluid were measured by enzyme linked immunosorbent assay.Combined with the evaluation index of patients ART treatment,statistical knowledge was used to analyze whether there was oxidative imbalance in follicular fluid of EMs patients,and whether the expression levels of oxidative stress markers SOD and AOPP were correlated with ART treatment.Results1.Comparison of general data between the two groups: there was no significant difference in age,BMI,years of infertility and serum b FSH,b LH and b E2 between the two groups(P>0.05).2.Comparison of evaluation indexes of ART treatment: the egg acquisition rate of the EMs group was significantly lower than that of the control group in the evaluation indexes of ART treatment of the two groups(P<0.05),There was no significant difference between the two groups in MII egg rate,fertilization rate,cleavage rate and excellent embryo rate(P>0.05).3.Comparison of oxidative stress markers SOD and AOPP in follicular fluid: the content of SOD in follicular fluid in EMs group was significantly lower than that in control group,and the difference was statistically significant(P < 0.05).There was no significant difference in the content of AOPP in follicular fluid between EMs group and control group(P >0.05).4.Correlation analysis between SOD,AOPP in follicular fluid and evaluation indexes of ART treatment: there was a poor positive correlation between SOD content in follicular fluid and egg acquisition rate(r = 0.261,P < 0.05),but no correlation with MII egg rate,fertilization rate,cleavage rate and excellent embryo rate(P > 0.05).The content of AOPP in follicular fluid was moderately negatively correlated with fertilization rate(r=-0.334,P < 0.05),but not with egg acquisition rate,MII egg rate,cleavage rate and excellent embryo rate(P > 0.05).Conclusions1.In this experiment,there was no significant difference in the content of AOPP in follicular fluid between EMs group and control group,but the content of SOD in follicular fluid of EMs patients decreased significantly,suggesting that there may be an oxidative imbalance in follicular fluid.2.There is a positive correlation between SOD content in follicular fluid and egg acquisition rate,and a negative correlation between AOPP content and fertilization rate,suggesting that SOD content and AOPP content in follicular fluid may be related to egg acquisition rate and fertilization rate.3.In this experiment,the content of SOD in follicular fluid in EMs group decreased significantly,and there was a positive correlation between SOD content and egg acquisition rate,suggesting that improving the level of oxidative stress in follicular fluid may provide a new idea for improving the outcome of ART treatment in patients with EMs infertility. |
