| Objective:Looking for the influencing factors of the hospitalization time after surgery in children with CHD after surgery.Then analyze the difference of gene expression profiles between children with Congenital Biliary Dilatation(CBD)and healthy children.Methods:We analysis the data of 66 children diagnosed with CBD after laparoscopic surgery in our hospital.Then we screen difference expression genes by c DNA microarray to detect and compare gene expression in children with CBD and healthy children.Analysis differential genes by GO and Pathway and verify differential expression genes related to inflammatory by q RT-PCR.Results:1.The factors influencing the the hospitalization time after surgery in children with CBD include white blood cells and total bilirubin at admission.Finally,we find the independent risk factor for the hospitalization time after surgery in children with CBD is white blood cell values at admission.2.There were 696 differentially expressed genes(cutoff =2.0)which 301 were upregulated and 395 were downregulated in children with CBD.Significant differences in gene expression profiles between children with CBD and healthy children has showed in cluster maps;Significant differences expressed genes in biological processes and molecular functions also has showed in GO analysis.The down-regulated pathways were mainly tumor necrosis factor signaling pathway,NF-kappa B signaling pathway,etc.IL1 B,IL1RN,TNF,TNFAIPA3 expression trends were consistent with chip results verified by q RT-PCR.Conclusion:1.The presence of infection on admission may affect the length of the hospitalization time after surgery in children with CBD.2.There are different Gene expression profile between children with CBD and healthy children.IL1 B,IL1RN,TNF,TNFAIPA3 were down-regulated in children with CBD. |
