| In recent years,the large-scale outbreak of infectious diseases,especially COIVD-19,has seriously threatened human’s life and health,which makes a deeply serious impact on the society and human’s property.Therefore,timely diagnosis and treatment of infectious diseases are of great significance in preventing the great spread of the infectious diseases.As the gold standard of etiological diagnosis,nucleic acid molecular diagnostic technology plays an increasingly important role on the prevention and control of infectious diseases.The rapid,lowcost,flexible,convenient and automatic nucleic acid sample processing platform is crucial for the effective application of nucleic acid molecular diagnostic.However,traditional diagnostic instruments with certain modules,single functions and inability of expanding can’t meet the demands for biosafety.Thence,taking advantages of open-source hardware on prototyping sample processing platform rapidly,an automated desktop nucleic acid samples processing platform based on the open source has been developed in this research.It can integrate the module of nucleic acid extraction and purification and the module of pipetting and gradient dilution efficiently.Meanwhile,through this platform,high-ratio gradient dilution can be accomplished for digital LAMP analysis.According to verification results,the platform can achieve nucleic acid extraction from which’s least concentration is 0.63 ng/μL,accurate 1 μL pipetting and 10-fold,100-fold,1000-fold high-rate dilution.It can deal with the demands of regions with relatively backward resources for efficient and low-cost rapid prototyping different molecular diagnostic technology services.In this research,the key innovation reflects in following three aspects:1.A low-cost,automatic rapid prototyping solution of nucleic acid sample processing based on open source hardware is proposed.The module of nucleic acid extraction and purification and the module of pipetting and gradient dilution are designed,which can be switched and expanded according to the needs of following molecular diagnosis.2.The hydraulic pipetting scheme is designed and can complete high-ratio gradient dilution to meet the needs of automatic and accurate pipetting for detection.3.The module of pipetting and gradient dilution is applied with digital droplet loop-mediated isothermal amplification(LAMP).The method is established to verify the function of high gradient dilution for nucleic acid samples with digital LAMP. |
