Mechanism Of Guizhi Fuling Pill On Pathological Angiogenesis In Rats With Hepatic Fibrosis

Purpose:In this study,the rat model of liver fibrosis was induced by carbon tetrachloride and Guizhi Fuling pills(GFW)with different doses were used to intervene the model rats,to observe the preventive and therapeutic effect of GFW on the model rats,and to study the effect of GFW on the expression of VEGF and VEGFR-2 gene in the liver tissue of model rats,to explore its anti-angiogenesis mechanism in the process of liver fibrosis with experimental methods,and provide new ideas and scientific basis for the clinical application of GFW in the prevention and treatment of liver fibrosis.Content:60 male SD rats were randomly divided into 6 groups: normal control group,model control group,Guizhi Fuling pill low、medium、high dose group,Dahuang Zhe Chong pill group.Except for the normal control group,the other groups were intraperitoneally injected with 40% CCl4 mixture twice a week to establish the rats’ liver fibrosis model.At the same time,rats in each group began to gavage,Guizhi Fuling pill low、medium、high dose group were given Guizhi Fuling pill solution of low、medium、high dose,Dahuang Zhe Chong pill group were given Dahuang Zhe Chong pill solution,normal control group and Dahuang Zhe Chong pill group were given the same amount of normal saline.After 8 weeks,the rats were killed to observe the pathological morphology of liver tissue,detect the serum liver function,HA content,CD34,VEGF,and VEGFR-2 gene expression.Results:1.GFW can downregulate the liver index of liver fibrosis model rats;2.GFW can reduce the IOD value of liver tissue in liver fibrosis model rats;3.GFW can reduce the contents of ALT,AST,TBIL,and HA in serum,and raise the level of ALB in serum of liver fibrosis model rats;4.GFW can inhibit the expression of CD34,VEGF,and VEGFR-2 m RNA in liver fibrosis model rats.Conclusion:1.GFW can reduce the pathological changes of liver cells and collagen fiber deposition,and reduce serum HA in liver fibrosis model rats,which suggests that GFW has certain anti-fibrosis effects;2.GFW can reduce the contents of ALT,AST,TBIL,and raise the level of ALB in serum of model rats,suggesting that GFW has a certain protective effect on liver function;3.The mechanism of GFW on liver fibrosis may be through affecting the signal of VEGF / VEGFR-2,inhibiting the m RNA expression of VEGF and VEGFR-2,and preventing the proliferation of pathological blood vessels.
