The degradation of physiological functions and disease appear during the aging.People start to pay more attention to delay aging.Oxidative stress plays a crucial role in accelerating aging and inducing neurodegenerative disease.The drug can effectively delay aging and prevent aging-related diseases.Red deer(Cervus elaphus)antlers contain avariety of bioactive ingredients,while its effects on anti-aging and aging-related diseases have not been reported yet.The present study is aim to explore the antioxidant and anti-aging activity of red deer antler water extract and its potential mechanism by the Caenorhabditis elegans(C.elegans).The results are as follow:(1)The antioxidant activity of red deer antler water extractOxidative stress model was used to evaluate the antioxidant effects of red antler water extract on the wild-type worms at different concentrations.The result showed that 1mg/m L the red deer antler water extract significantly exerted anti-oxidant activity in vivo and prolonged the survival time under oxidative stress conditions compared with others concentrations.(2)The anti-aging activity of red deer antler water extractLifespan test,stress test(UV/heat)and physiological function test(swallow/swim/generate)were used to explore the effect of 1mg/m L red deer antlers water extract on the senescence of wild-type worms.The results showed that the red deer antlers water extract could significantly prolong the survival time of wild-type nematode(P<0.01)and improve the ability of nematode to UV and high temperature stress(P<0.01),swallowing frequency(P<0.01),number of swings and egg production(P<0.01).It showed significantly anti-aging activity.(3)The protective effect of red deer antler water extract on the Aβ-induced paralysisThe experiment of Aβ-induced paralysis was used to detect the protection of the water extract from red deer antler on transgenic nematode CL4176.The experimental results showed that the red deer antler water extract could significantly delay the paralysis induced by Aβ.(4)The mechanism of anti-aging biological activity of red deer antler water extractThe experiment of ROS level was used to evaluate the effect of water extract from red deer antlers on reactive oxygen species(ROS)in nematodes.The experiments of oxidative stress and lifespan were used to explore the effects of mitochondrial and SKN-1 on anti-aging activity of red deer antler water extract in mutant nematodes clk-1(qm30)and skn-1(zu35).The effects of the water extract from red deer antlers on the transcription of classic anti-oxidant genes in nematodes were detected by the experiment of q-PCR.The red deer antler water extract could significantly reduce the level of reactive oxygen species in nematodes,but had no significant effects on the antioxidant activity and lifespan of mutant nematodes clk-1(qm30)and skn-1(zu35),and could significantly improve transcription level of classic antioxidant genes which were downstream of transcription factor SKN-1,such as gst-4,gst-7.In summary,the anti-aging effect of red deer antler water extract was regulated by the function of mitochondrial.The water extract from red deer antler could up-regulate the transcription level of antioxidant genes downstream of skn-1 when the pathway of SKN-1/Nrf2 was activated.It could also enhance the antioxidant ability of nematodes,reduce the damage of oxidative stress,delay the senescence of nematodes.
