| Aging is usually characterized by a decrease in the body’s ability to adapt physically and psychologically to changes in the environment.The incidence of chronic diseases such as cancer,diabetes and cardiovascular disease increases with age.Population aging poses a great threat to international medical system and economic development.Drug therapy can effectively relieve aging,but long-term use of antiaging drugs such as aspirin and metformin will cause adverse reactions such as antiplatelet aggregation,diarrhea,nausea and abdominal pain.Therefore,it is urgent to develop safe and effective anti-aging methods.TCM has a wide range of clinical application value,such as the treatment of COVID-19,cancer and anti-aging.Among them,Honeysuckle,Polygonum multiflorum,glycyrrhiza,Lycium barbarum polysaccharide and Astragaloside IV have significant anti-aging activities.Ginseng(Panax ginseng C.A.Meyer),is A kind of extremely precious Chinese herbal medicine,has the effect of reducing oxidative stress,anti-aging and preventing senile dementia.As the main component of ginseng body(about 0.1-0.5%),ginseng volatile oil has been reported to have anti-inflammatory and anti-tumor activities,but whether it can delay aging has not been reported.Using Caenorhabditis elegans as a model,this study investigated the anti-aging effect of ginseng volatile oil and its possible mechanism.The specific research contents are as follows:1.The essential oil of ginseng was obtained by supercritical CO2 extraction and detected by Gas chromatography-mass spectrometry(GC-MS).A total of 73 substances were detected.The main ingredients were ginseng alkynyl alcohol(17.51 %)and ginseng epoxynyl alcohol(17.15 %).2.The effects of ginseng volatile oil on the longevity and survival rate of wild-type c aenorhabditis elegans(N2)after heat stress were observed by longevity test and he at stress test.It was found that ginseng volatile oil increased the life span and survi val rate of nematodes after heat stress.3.To observe the effects of ginseng volatile oil on senescence related physiological i ndexes of nematodes,including motor capacity,reproductive capacity,body lengt h and lipofuscin level.The results showed that panax ginseng volatile oil could im prove the locomotion ability,increase the body length and decrease the lipofuscin level without damaging the reproductive function of nematodes.4.DPPH free radical scavenging ability of ginseng volatile oil was detected in vitro,and the effects of ginseng volatile oil on SOD activity,MDA content and ROS lev el of nematodes in vivo were determined.The results showed that ginseng volatile oil had strong scavenging ability of DPPH free radical,and could increase SOD a ctivity,decrease MDA content and ROS level in nematodes.5.Transcriptome sequencing was used to extensively identify the effects of ginseng volatile oil on nematode gene levels.Results a total of 3139 up-regulated genes an d 3190 down-regulated genes were identified,and the differentially expressed gen es were significantly enriched in peroxidase,autophagy and nucleotide extraneous repair pathways.6.To observe the effects of ginseng volatile oil on the longevity of SOD-1 mutant ne matodes and the fluorescence intensity of P62 protein in BC12921 nematodes.Th e results showed that ginseng volatile oil significantly reduced the fluorescence int ensity of P62 protein in BC12921 nematodes,but did not prolong the lifespan of S OD-1 mutant nematodes.In conclusion,ginseng volatile oil can prolong the life span and healthy life span of C.elegans without damaging its reproductive capacity,and its life-prolonging activ ity is closely related to antioxidant and autophagy pathways. |
