Objective:In this study,the difference of TOMM40 protein expression level in colorectal cancer tissues and paired adjacent tissues was explored by bioinformatics database,2d liquid chromatography-mass spectrometry and immunohistochemistry,and the effect of TOMM40 protein expression level on biological function of colorectal cancer cells was further verified by cell function experiment.It provides a new direction for the diagnosis and treatment of colorectal cancer and another possibility for exploring the pathogenesis of colorectal cancer.Methods:1.The expression of TOMM40 in colorectal cancer was determined by referring to Timer database and GEPIA database,and the co-expressed genes of TOMM40 were obtained by Cbioportal database.2.The expression of TOMM40 protein in 16 pairs of colorectal cancer and its adjacent tissues was detected by 2D liquid chromatography-mass spectrometry,and the results were verified by Western blot.Immunohistochemistry was used to analyze clinical tissue samples,and TOMM40 protein expression was detected in 40 pairs of colorectal cancer tissues and their adjacent tissues.3.SW620 colorectal cancer cell line was selected as the target cell line to construct TOMM40 overexpression and silenced expression cell lines,which were verified by fluorescence microscopy,RT-QPCR and Western Blot test.Then the effects of TOMM40 protein expression on cell biological function were detected by cell proliferation assay,cell migration and invasion assay,and cell apoptosis assay.Results:1.TOMM40 is up-regulated in colorectal cancer tissues.2.The up-regulated expression of TOMM40 in colorectal cancer tissues was verified by 2D liquid chromatography-mass spectrometry and Western Blot.Immunohistochemical analysis of clinical tissue samples also confirmed the upregulated expression of TOMM40 in colorectal cancer tissues.3.Transient transfection SW620 overexpression and silencing expression cell lines were successfully constructed.Cell proliferation and migration and invasion experiments showed that overexpression of TOMM40 could promote cell proliferation,migration and invasion,while silencing expression of TOMM40 could inhibit cell proliferation,migration and invasion.Apoptosis assay showed that overexpression of TOMM40 inhibited apoptosis and silencing expression of TOMM40 promoted apoptosis.Conclusions:1.TOMM40 is up-regulated in colorectal cancer tissues.2.The up-regulated expression of TOMM40 can promote the proliferation,migration and invasion functions of colorectal cancer cells,and the up-regulated expression of TOMM40 can inhibit the apoptosis of colorectal cancer cells.TOMM40 silencing can inhibit the proliferation,migration and invasion of colorectal cancer cells and promote the apoptosis of colorectal cancer cells.3.TOMM40 has potential as a biomarker for the diagnosis and treatment of colorectal cancer.
