The Differential Expression And Biological Function Of LMCD1 In Colorectal Cancer | | Posted on:2024-02-23 | Degree:Master | Type:Thesis | | Country:China | Candidate:C R Zhao | Full Text:PDF | | GTID:2544307064987899 | Subject:Clinical Laboratory Science | | Abstract/Summary: | PDF Full Text Request | | Objective:In this study,the differential expression of LMCD1 protein in colorectal cancer tissues and paracancer tissues was studied by bioinformation technology,two-dimensional liquid chromatochromatoid-mass spectrometry,Western blot and immunohistochemistry,and the biological function of the differential expression of LMCD1 protein in colorectal cancer cells was further verified by cell function experiments.It provides a new target in the early screening of colorectal cancer and a new strategic idea in the investigation of the biological mechanism of colorectal cancer.Methods:1.The Timer database was used to determine the differential expression of LMCD1 in colorectal cancer.The gene expression profile data of normal colorectal tissues and colorectal cancer tissues were mined through the TCGA database,and the expression of LMCD1 in colorectal cancer was confirmed again through statistical calculation.The association between LMCD1 and cancer immune subtypes was found in the TISIDB database.Gene mutation and co-expression of LMCD1 were mined from Cbioportal database.Heat maps and functional enrichment maps of LMCD1 positive and negative related genes were obtained through Linked Omics database.The interaction between LMCD1 proteins was explored through STRING database.2.The expression difference of protein LMCD1 in 24 cases of colorectal cancer and paracancer tissues was detected by two-dimensional liquid chromatogen-mass spectrometry.The protein expression of LMCD1 in wax blocks of colorectal cancer and its adjacent tissues was determined by immunohistochemistry.3.SW620 cell lines with LMCD1 overexpression and silent expression were successfully constructed.The transfection efficiency was verified by green fluorescence,RT-q PCR and Western blot.The effects of differential expression of LMCD1 protein on cell biological function were detected by cell proliferation assay,cell scar assay.Results:1.The expression of LMCD1 is up-regulated in colorectal cancer and other tumors.2.The upregulated expression of LMCD1 in colorectal tissues was demonstrated by two-dimensional liquid chromatography and mass spectrometry,which was also verified by western blotting and immunohistochemistry.3.Transient transfected SW620 overexpression and knockdown expression cell lines were successfully constructed.Cell function experiments showed that overexpression of LMCD1 could promote cell proliferation and migration,while silence expression of LMCD1 could inhibit cell proliferation and migration.Conclusions:1.LMCD1 is up-regulated in colorectal cancer tissues.2.The up-regulated expression of LMCD1 can promote the proliferation and migration function of colorectal cancer cells,and may be used as a potential biomarker for screening and diagnosis of colorectal cancer. | | Keywords/Search Tags: | LMCD1, colorectal cancer, Bioinformatics, two-dimensional chromatography and mass spectrometry, immunohistochemistry, cell function experiment | PDF Full Text Request | Related items |
