Objective:To investigate the expression of KDM5A,E-cadherin and Vimentin in colorectal cancer and adjacent normal tissues,and analyze the relationship between the expression of KDM5A,E-cadherin and Vimentin in CRC and the clinicopathological features of patients.To explore the role of KDM5A in CRC and its relationship with EMT.Methods:From July 2021 to December 2021,56 cases of tumor tissue specimens and normal tissues adjacent to cancer were collected from the Department of Pathology of Affiliated Hospital of Yan’an University.The expressions of KDM5A,E-cadherin and Vimentin were detected by IHC,and the experimental data were analyzed by statistical methods.Results:1.KDM5A was positive in CRC in 47 cases(83.9%),and in normal tissues adjacent to cancer in 15 cases(26.8%).The positive expression of KDM5A in CRC was higher than that in normal tissues adjacent to cancer,and the difference was statistically significant(χ~2=39.510,P<0.001).2.The expression of 2.KDM5A was related to lymph node metastasis(χ~2=6.093,P=0.014),differentiation degree(χ~2=8.023,P=0.005)and clinical stage(χ~2=4.274,P=0.039),and the difference was statistically significant.There was no significant difference in age(χ~2=0.996,P=0.318),sex(χ~2=0.000,P=1.000),distant metastasis(P=0.415)and infiltration depth(2=0.000,P=1.000).3.Vimentin was positive in CRC in 10 cases,with a positive rate of 17.9%(10/56).The normal tissues adjacent to cancer were all negative(0/56).The positive expression of vimentin in adjacent normal tissues was lower than that in tumor tissues,and the difference was statistically significant(χ~2=10.000,P=0.001)。The positive rate of E-cadherin in CRC was 83.9%(47/56).There was no loss of expression in normal tissues adjacent to cancer,and all of them were positive(56/56).The negative expression of E-cadherin in CRC was higher than that in adjacent tissues,and the difference was statistically significant(χ~2=10.980,P=0.001).4.The expressions of E-Cadherin and Vimentin were related to the degree of tumor differentiation(χ~2=4.329,7.742,P=0.037,0.005),and the difference was statistically significant.Age(χ~2=0.000,P=1.000),sex(χ~2=0.000,P=1.000),distant metastasis(P=1.000),infiltration depth(χ~2=1.348,1.654,P=0.246,0.198),Clinical stage(χ~2=2.475,0.225,P=0.116,0.636)and lymph node metastasis(χ~2=3.772,0.733,P=0.052,0.392)had no significant difference.5.The expression of E-cadherin was negatively correlated with Vimentin(rs=-0.558,P<0.001).6.The expression of KDM5A protein was correlated with E-cadherin and Vimentin,but there was no statistical significance(rs=-0.179,0.204,P=0.185,0.131).Conclusion:1.The high expression of KDM5A in CRC is related to the clinical stage,differentiation degree and lymph node metastasis of patients.It is speculated that the high expression of KDM5A may be related to the poor prognosis of CRC.2.The results of E-cadherin and Vimentin indicate that EMT may be related to the occurrence and development of CRC.3.The expression of KDM5A was related to E-cadherin and Vimentin,but there was no significant difference.The relationship between KDM5A and EMT needs futher study.
