Objective:Our aim is to observe the therapeutic effect of Lycium barbarum polysaccharide-glycoprotein(Lb Gp)in Amyotrophic lateral sclerosis and to initially explore its possible mechanisms.Method:1.SOD1G93Atransgenic female mice were randomly divided into saline group(TG-NS,n=16)and Lycium barbarum polysaccharide-glycoprotein group(TG-Lb Gp,n=16).At the same time,the age and sex matching wild-type female mice were randomly divided into saline group(WT-NS,n=16)and Lycium barbarum polysaccharide-glycoprotein group(WT-Lb Gp,n=16).Starting from 90 days of age,SOD1G93Amice were given Lb Gp(20 mg/kg)once daily by intragastric administration until death.2.0bserved the effects of Lb Gp on the body weight,disease onset,lifespan,and disease duration in SOD1G93Amice.3.Immunofluorescence staining of spinal cord to assess the effect of Lb Gp on astrocytes.4.Evaluate the activation and phenotypic changes of microglia in spinal with immunofluorescence and q PCR.5.Use q PCR to detect changes in the relative expression of inflammatory factors in the spinal cord.Results:Our data suggested that repeated Lb Gp administration prolonged the lifespan,extended disease duration of SOD1G93Amice by about 17.5%and 35.1%,respectively.Mechanistic studies have shown that Lb Gp inhibit neuroinflammation by inhibiting astrocyte and microglia activation,regulating the phenotype of microglia,inhibiting their polarization to M1 phenotype,and promoting their polarization to M2phenotype.Conclusion:Lb Gp may become a potentially effective treatment for ALS.
