Objective: To study the expression difference spectrum of plasma exosomes mirnas in patients with gastrointestinal stromal tumor and normal controls,and screen out specific mi RNAs in plasma exosomes of patients with stromal tumor,providing a theoretical basis for further study on the biological characteristics of plasma exosomes mi RNAs in patients with stromal tumor.Methods:From June 2020 to June 2021,5 patients with mesenchymal tumor and 5healthy people who underwent physical examination were selected as research objects.Mi RNAs in plasma exosomes of stromal tumor patients and healthy people are extracted,and the mi RNAs in exosomes are detected by high-throughput RNA sequencing method and compared with RNA libraries.Screen out significant differences of mi RNAs(p < 0.05 and | log2 FC | > 1).Results:Compared with normal controls,the expression of plasma exosome mi RNAs in patients with gastrointestinal stromal tumor is different,and the number of differentially expressed mi RNAs is 302,among which 150 are up-regulated and 152 are down-regulated.mi RNA-148a-3p is up-regulated in the exosomes of patients with stromal tumor.Conclusions:There are significant differences in the mi RNAs in plasma exosomes of patients with gastrointestinal stromal tumor compared with normal controls,suggesting that the exosomal mi RNAs of patients with gastrointestinal stromal tumor have specificity,and may have potential research value in the diagnosis and treatment of stromal tumor.It is found that exosomal mi RNA-148a-3p increased significantly in stromal tumors,which may be involved in the biological process of stromal tumors and may be a marker for diagnosis and prognosis.Objective: To detect the expression of miRNA-148a-3p in plasma exosomes of patients with stromal tumor and normal controls by q RT-PCR,and to study the relationship between mi RNA-148a-3p and clinical data.Methods:A total of 37 patients with stromal tumor and 10 healthy people admitted to our hospital from June 2020 to June 2021 were selected as the research objects and divided into the patients with stromal tumor and the healthy group.Exosomes in plasma of the patients and healthy people are isolated by ultra-fast centrifugation method,and total exosomes RNA is extracted.The expression of mi RNA-148a-3p in the exosomes of the stromal tumor group and the normal control group is quantitatively detected by q RT-PCR,and the clinical data of the patients with stromal tumor,including gender,age,tumor size,mitotic image and risk,are collected to analyze the relationship between mi RNA-148a-3p expression and clinical data.Results:The expression level of mi RNA-148a-3p in plasma exosomes of patients with gastrointestinal stromal tumor is significantly higher than that of normal controls(about4.43 times),and its high expression is not related to gender or age(P > 0.05),but positively correlated with tumor size,mitotic figure and risk(P > 0.05).Conclusions : The expression of mi RNA-148a-3p in the exosomes of patients with gastrointestinal stromal tumor is significantly higher than that of normal controls,and the expression of mi RNA-148a-3p is different(P > 0.05),and the differentially expressed mi RNA-148a-3p is related to the factors of poor prognosis of stromal tumor.It is suggested that exosomal mi RNA-148a-3p may be a marker for diagnosis and prognosis of stromal tumor.
