Objective:Cervical cancer ranks fourth among female malignant tumors in terms of morbidity and mortality worldwide.More than 80 percent of women will be infected with HPV during their lifetime,but most of the time HPV can be cleared by an immune mechanism that is not well understood,and if HPV persists,it can eventually lead to cervical lesions.The stability and composition of the vaginal microbiome may play an important role in determining the host’s innate immune response and susceptibility to infection.But little is known about the mechanisms associated with clearance or persistence of infection.Before most of the studies on vaginal microecological all depends on the development of methods to assess the microbial community composition,however under the vaginal microenvironment,the number of anaerobic bacteria and aerobic bacteria can’t develop so about the detection of micro ecological environment there are many uncertain factors,the development of 16 s r DNA technology,can enable us to more direct,more rapid analysis of microbial groups.In this study,16 S r DNA sequencing technology was used to compare and analyze the vaginal microflora of hp V-16,HPV-52 and HPV-negative women with no cervical lesions treated in the First Affiliated Hospital of Dali University,so as to reveal the changes of vaginal microecology in patients with different TYPES of HPV infection.Thus it provides a theoretical basis for more accurate personalized medical treatment for patients with different infection types by adjusting vaginal microenvironment.Method:Women who were treated in the first affiliated Hospital of Dali University from June 2021 to January 2022 were selected according to our inclusion and exclusion criteria.Under the condition of informed consent and written informed consent,routine leucorrhea examination,HPV typing and TCT examination were performed during gynecological examination,and vaginal secretions were taken and stored in-80 °C refrigerator with sterile cotton swab at vaginal fornix.Finally,4 patients with HPV16 infection without cervical disease(group N16),4 patients with HPV52 infection without cervical disease(group N52),and 4 patients without HPV infection without cervical disease(group C)were selected.The characteristics of vaginal flora in 12 samples were analyzed by 16 sRNA.Results1.Microbial species diversity was significantly decreased in the generic HPV infection group(N16 group and N52 group).And the abundance of Lactobacillus increased significantly after HPV infection.The bacteria with the highest sample abundance in the three groups were lactobacillus and all of them were > 50%.After HPV infection(N16 group,N52 group),the abundance of Lactobacillus increased significantly,while the abundance of Prevotella,Gardnerella,Ciliomycetes,Strangella,Megalococcus and Dialysteria decreased.2.After infection with HPV16 and 52,From bronze bacteria genera,staphylococcus aureus,dynamic bending coli,gardnerella,slaughter bacterium bacillus genus,dung,only cell bacteria genera,Sartre’s bacteria genera,streptococcus,leptothrix,Bulleidia,digestive streptococcus,small single cell bacteria,staphylococcus aureus,giant spherical bacteria,fusobacterium,exotic species,Wei Rong bacteria,platts bacteria genera bacteria of the genus listeria,wear Decreased abundance.3.HPV16 infection-specific bacteria: Rothia,red coli,lactobacillus,burkholderia bacteria genera,bacillus,gathered lamb bacteria,bifidobacterium,aquatic bacteria genera,staphylococcus aureus,enterococcus,mycoplasma,anaerobic bacteria genera,puzzled sugar,peptone,porphyrin genera,Alloscardovia,fecal bacteria bacterium,bacteroides,campylobacter,fingal DE bacteria genera,WAL1 855 D,1-68.4.The specific bacteria of HPV52 infection are Prosella,Delfteia,Phyllobacter,Acinetobacter,Ureaplasma,Pseudomonas,Bacillus,Shigella,Blastomycetes,Roxella,Red bacillus,Lactobacillus and Burkholella.Conclusion1.HPV infection caused significant changes in the microecology of vaginal flora;2.There are differences in the changes of vaginal flora caused by HPV type 52 and 16 infection.The degree of vaginal microecological changes caused by type 52 infection was more complicated than that caused by type 16 infection.
