Objective:To explore the expression and significance of TGF-β1 and E-cadherin in endometrial carcinoma and normal endometrial carcinoma,and analyze the effect of their expression on the prognosis of patients,to further study the correlation difference with the expression of HOXB9 in endometrial carcinoma,and to analyze the mechanism of HOXB9 in the progression of endometrial carcinoma.Methods:171 cases of endometrial carcinoma tissues and 35 normal endometrial specimens were collected from the First Hospital of Lanzhou University,Gansu Cancer Hospital and Gansu Maternal and Child Hospital.The expression of TGF-β1and E-cadherin was detected by tissue microarray and immunohistochemistry.The expression differences of HOXB9,TGF-β1 and E-cadherinin EC and normal endometrial were analyzed byX~2test and their relationship with clinicopathological features.The correlation of HOXB9,TGF-β1 and E-cadherin in EC was analyzed by SPSS statistical software.Kaplan-meier log-rank test was used to analyze the effect of TGF-β1 and E-cadherin expression levels on the overall survival of patients with EC,and the survival curve was drawn.Cox multivariate regression was used to analyze the prognostic factors of EC.Result:1.We examined TGF-β1 expression in 171 cases of endometrial carcinoma tissue samples using immunohistochemical staining and compared that to paired normal endometrial samples.The positive rate of TGF-β1 in endometrial carcinoma was33.9%(58/171),higher than the rate of normal endometrium 5.7%(2/35),the difference was statistically significant(P<0.05);What’s more,in stage I or II endometrial carcinoma,the positive rate is 30.5%(43/141),which is significantly lower than stage III or IV endometrial carcinoma(50.0%,15/30);Besides,the positive rate of TGF-β1 in cancer tissues without lymph node metastasis was 30.7%,lower than carcinoma tissues with lymph node metastasis(30.7%,46/150)(P<0.05).There was no significant difference in the expression of TGF-β1 protein in menstrual status,histological grade,intravascular tumor plug,muscle layer invasion depth,recurrence and metastasis;2.Positive E-cadherin expression was found to be located in the cell membrane by immunohistochemical staining,with the positive rate of 47.9%(82/171)in cancer tissues,which is much lower than that in 35 controls(88.6%,31/35),the difference between the two groups was statistically significant(P<0.05).The expression rates of E-cadherin in type I and II EC were 53.7%and 34.0%,respectively,the difference was statistically significant(P<0.05);The positive rate of E-cadherin varies in EC tissues with varying degrees of differentiation,in the endometrial cancers of the histological grade G1,G2,and G3 groups,the expression rate of E-cadherin was66.0%(31/47),54.1%(20/37),and 35.6%(31/87),respectively,The difference was statistically significant(P<0.05),In terms of the clinical staging,the positive expression rate of E-cadherin was 52.5%in 141 endometrial cancer tissues with stage I or stage II(74/141),the expression rate in 30 stage III or IV EC tissues was 26.7%(8/30),the difference was statistically significant(P<0.05);In terms of myometrial infiltration,among the 96 EC tissues with infiltration depth≤1/2 myometrium,the positive rate of E-ca was 55.2%(53/96),higher than the positive rate in 75 patients with depth>1/2 myometrium(38.7%,29/75),this difference was statistically significant(P<0.05);There was no significant difference in E-cadherin protein expression in menstrual status,intravascular cancer thrombolysis,postoperative chemo-chemotherapy,lymph node metastasis,recurrence and metastasis;3.The expressions of HOXB9 and TGF-β1 proteins were positively correlated in endometrial carcinoma tissues,but the expressions of HOXB9 and TGF-β1 protein were negatively correlated with the expression of E-cadherin in endometrial carcinoma tissues;4.Univariate analysis showed that the expression levels of TGF-β1 and E-ca in EC not affected the overall survival of patients.Multivariate analysis showed that TGF-β1 and E-cadherin expression had no effect on the prognosis of EC patients.Conclusion:1.TGF-β1 and E-ca may play an important role in the progression of endometrial carcinoma;2.TGF-β1 positively promoted the progression and metastasis of endometrial carcinoma;E-ca also plays an important role in endometrial carcinoma invasion.3.HOXB9,TGF-β1 and E-ca have a certain mutual regulatory effect on the progression of endometrial cancer,and HOXB9 and TGF-β1 May synergistically participate in the invasion and metastasis of endometrial cancer.
