Effect Of Houpu Sanwu Decoction On Interstitial Cells Of Cajal And Receptor NK-1 In The Gastrointestinal Tract Of Rats With Disorders Of Gastro Intestinal Motility Diseases

Objective:By constructing a rat model of gastrointestinal motility disorder(DGIMD),we observed the effects of Houpu Sanwu Decoction on the expression of Cajal interstitial cells(ICCs)and neurokinin 1 receptor(NK-1)in the gastrointestinal tract of DGIMD rats and explored the mechanisms related to its effects on gastrointestinal motility,to provide more theoretical basis for the treatment of DGIMD with Houpu Sanwu Decoction.Methods:24 healthy adult male SD rats were randomly divided into the blank group(CG),model group(MG),Houpu Sanwu Decoction group(HPG),and Domperidone group(DG).(1)Molding: L-arginine(L-Arg)was used for intraperitoneal injection of molds.On day 1,MG,HPG,and DG rats were given 20% L-Arg solution intraperitoneally at 5.2 g/Kg according to body weight.From day 2 to day 5,MG,HPG,and DG rats were given 20% L-Arg solution intraperitoneally at 2.6 g/Kg according to body weight.CG rats were given the same dose of saline intraperitoneally during the modeling period.(2)Drug administration treatment: From day 6 to day 12,HPG and DG rats were given Houpu Sanwu Decoction(4.6g/Kg)and Domperidone suspension(6.3g/Kg)by gavage,and CG and MG rats were given saline gavage.(3)Sampling and testing: During the experimental period,we observed the weight,mental activity,hair color,and stool changes of each group of rats before,after,and after the treatment.The rats were given fasting treatment without water for 24 h at the end of treatment on day12,after which the rats were taken on the ice.The gastric emptying rate(GER)and small intestine advancement rate(SIPR)of each group were also observed.The expression of specific proteins stem cell growth factor(C-kit)and its ligand stem cell growth factor(SCF),neurokinin 1(NK-1),and its ligand substance P(SP)were detected in ICCs by enzyme-linked immunosorbent assay(ELISA),protein immunoblot(Western Blot)and immunohistochemistry(IHC).Results:1.From day 1 to day 5 of the modeling process,compared with CG rats,MG,HPG,and DG rats showed significant weight loss,depression,decreased activity,and loose and unformed stools.MG rats did not show significant weight gain during the drug treatment from day 6 to day 12.In contrast,HPG and DG rats gradually gained weight,gradually increased activity,and formed stools after drug gavage.2.Compared with CG rats,GER and SIPR were significantly decreased(P < 0.01),and serum MTL level was reduced considerably(P < 0.01)in MG rats.Compared with MG rats,GER and SIPR were significantly increased in HPG and DG rats(P < 0.01),and serum MTL level was significantly increased(P < 0.01).3.Compared with CG rats,gastric,jejunal,ileal,and colonic C-kit and its ligand SCF,NK-1,and ligand SP proteins were significantly decreased in MG rats(P < 0.01).In contrast,the expression of the above proteins was increased dramatically in HPG and DG rats(P < 0.01).4.Compared with HPG rats,there was no significant difference in the expression of gastric,jejunal,ileum,colon C-kit and its ligand SCF,NK-1 and its ligand SP protein in DG rats.Conclusions:1.The group demonstrated the feasibility of constructing DGIMD rats by intraperitoneal injection L-Arg method through repeated experiments.2.Houpu Sanwu Decoction can effectively improve the gastrointestinal motility of DGIMD rats and can promote weight gain.3.Decreased gastrointestinal motility in DGIMD rats was associated with SCF/Ckit signaling pathways in the gastrointestinal tract and down-regulation of NK-1 and ligand SP expression.4.The promoting effect of Houpu Sanwu Decoction on gastrointestinal motility may be related to the upregulation of SCF/C-kit signaling pathways in the gastrointestinal tract and the expression of NK-1 and ligand SP.
