High performance liquid chromatography(HPLC)has the advantages of high separation efficiency and stable performance,and plays an important role in many fields.By introducing specific groups into analytes,chemical derivatization can improve the performance of chromatographic analysis and greatly broaden the application range of HPLC.At present,most of the commercial fluorescent derivatives are based on coumarin,rhodamine and fluorescein,but the labeling reagents based on 1,8-naphthalimide parent are rarely reported.In this work,six fluorescent derivatives for labeling aldehydes and ketones were prepared from1,8-naphthalimide.A series of sensitive and accuracy HPLC methods for the determination of fatty aldehydes and aromatic aldehydes were developed by using these new reagents with fluorescence detection(FLD)or ultraviolet detection(UV),which provided methodological support for food and drug safety and quality control.The main contents are as follow:Chapter 1: the research progress of structural characteristics for naphthalimide,chemical derivatization and analytical reagents based on naphthalimide were reviewed.With the advantages of low cost,easy modification and excellent fluorescence,1,8-naphthalimide has been widely used in the field of sensing and imaging.However,studies as fluorescent derivatives have been rarely reported.Chapter 2: Design and prepared six pre-column derivatization reagents using1,8-naphthalimide as core structure,namely NPHNA,NBHNA,i-NBHNA,NPeHNA,i-NPeHNA,NCyHNA.The obtained six pre-column derivatization reagents were characterized by MS and NMR.Chapter 3: Using NBHNA as the pre-column derivatization reagent,a method for the accurate determination of 8 kinds of fatty aldehyde was established by HPLC-FLD.The method is applied for the determination of aldehydes in soybean oil.The method has a great significance of the determination of aldehyde in food.Chapter 4: Using NPeHNA and i-NPeHNA as analogue-coded derivatization reagents,A semi-targeted HPLC-FLD was developed to dynamically monitor aldehydes in deep-heated soybean oil.And the potential aldehyde markers were identified by MS.This work has important reference value for the screening of aldehydes quality control index,and can provide technical support for monitoring the quality and safety of edible oil.Chapter 5: Using NCyHNA as the pre-column derivatization reagent,an accurate and rapid method for the determination of three furfuralwas established by HPLC-FLD.The method was successfully used to detect three furfural analogues in raisin samples.This method is of great significance for the evaluation of furfural content in food.Chapter 6: Using NCyHNA as pre-column derivatization reagent,a new method for sensitive determination of genotoxic impurities 1,4-benzenedioxyhexacyl-6-aldehyde(BDA)and p-nitrobenzaldehyde(NBA)was established.This method has been successfully applied to the determination of aldehyde impurities in raw materials and preparations.The results show that the sensitivity and accuracy of the method can meet the requirements of drug impurity detection,and it is of great significance for drug quality evaluation and safe use.
