Objective: To prepare a rabbit animal model of hyperlipidemia by using exogenous high-fat diet induction method,use drug-separated cake moxibustion to intervene in hyperlipidemia rabbits,and use lipidomics as the starting point to explore the effect of drug-separated cake moxibustion on abnormalities The regulation of lipid metabolism provides a more substantial theoretical basis for the clinical application and promotion of drug-separated cake moxibustion in the treatment of hyperlipidemia.Methods: 36 healthy purebred male New Zealand white rabbits were randomly divided into 3 groups: normal group,model group,and medicated cake moxibustion.The hyperlipidemia rabbit model was prepared by the exogenous high-fat diet induction method.After the modeling was completed,the model group continued to be fed with the exogenous high-fat diet.The drug-separated cake moxibustion group was treated with the drug-separated cake moxibustion and normal diet intervention.The group did not receive any intervention measures.After the intervention,fasting,ear vein anesthesia,blood sampling from the abdominal aorta,sacrifice the animals,take liver tissues,observe the blood lipid levels of hyperlipidemia rabbits,and analyze by chromatography-mass spectrometry(LC-MS)The technique was used to explore the different metabolites of drug-separated cake moxibustion intervention in hyperlipidemia rabbits,and oil red O staining was used to observe lipid deposition in liver tissue.Result:1.Blood lipid level: After 8 weeks of intervention,compared with the model group,the serum TC,TG,LDL levels of the herbal cake-separated moxibustion group were significantly decreased(p<0.05);compared with the normal group,the serum TC,TG,LDL of the herbal cake-separated moxibustion group are still at a high level(p<0.05).2.Differential metabolites: The top 15 differential metabolites were screened out through LC-MS technology,which are PC 280,SM 52132 O,LPC 250/00,PC 366,SM 56132 O,SM 3202 O,PC 376,LPC O-240,SM3632 O,PC 424,LPC O-241,SM 3322 OSM 171,PC 323,PC 4210,PC344.3.Lipid deposition in liver tissue: Compared with the model group,the lipid droplets were reduced in the herbal cake-separated moxibustion group,and lipid deposition was significantly reduced;compared with the normal group,there were a small amount of lipid droplets in the herbal cake-separated moxibustion group.Conclusion:1.Herbal cake-separated moxibustion and diet control can significantly improve the blood lipid level of hyperlipidemia rabbits,and can effectively reduce lipid deposition in liver tissue;2.The top 15 differential metabolites,PC,SM and LPC,were screened out by lipomics,which belong to glycerophospholipids.After the treatment of herbal cake-separated moxibustion,the concentration of these metabolites decreased significantly.Therefore,the therapeutic mechanism of herbal cake-separated moxibustion may be related to the regulation of phospholipid metabolism,which needs to be further explored.
