Objective: To observe the changes of neurological deficit scores,serum cerebral ischemic area SYP/PSD-95 and its gene expression in MCAO rats with electro-acupuncture at the points of the pericardium meridian in MCAO rats,and to explore the effects of acupuncture at the points of the pericardium meridian on the synapses after cerebral ischemia The influence of plasticity provides theoretical support and experimental basis for clinical application of pericardium meridian points to treat cerebral ischemia.Method: This experiment adopts multiple random methods.For the first time,150 SD rats are randomly divided into 30 normal groups,30 sham-operated groups,and 90 models that need to be modeled;after the model is successfully modeled,90 models The rats were randomly divided into the model group,the pericardium meridian acupoint group,and the lung meridian acupoint group for the second time.Each group was randomly assigned to 5time points of 3d,7d,14 d,21d and 28 d.6 at each time point.The MCAO rat model was prepared by inserting a suture in the external carotid artery.The acupoints at different segments of the pericardium meridian and lung meridian were selected for electroacupuncture treatment on the second day after successful modeling.Each time,30 minutes,continuous electroacupuncture for 6 days at intervals 1 day.Observe the neurological deficit scores of rats,use ELISA,immunohistochemistry,and RT-q PCR to detect the expression of SYP,PSD-95,SYP,PSD-95 and m RNA in the cerebral ischemic area.Result:1.Neurological deficit score: comparison within the group: the score of the model group increased after cerebral ischemia injury,compared with 12 h,the score of the model group decreased significantly after 21 days(p<0.05,p<0.01),the pericardial meridian and lung meridian The group score decreased significantly after the 14 th day(p<0.01,p<0.05).Comparison between groups: Compared with the normal group and the sham operation group,the scores of the pericardial meridian group,lung meridian group,and model group at each time point were significantly increased High(p<0.05),and the pericardial meridian group and lung meridian group can down-regulate the neurological function score to varying degrees,but the difference is not statistically significant compared with the model group(p>0.05).2.SYP:(1)Serum SYP: intra-group comparison: group: 28d>21d>14d>7d>3d(p<0.01),the lung meridian group: 28d>21d>14d>7d(p<0.01,p<0.05),comparison between groups: the expression of the model group decreased at each time point,14,21 d,28d pericardial meridian group> lung meridian group> model group(p<0.01)And the role of the pericardium meridian group gradually increased over time.(3)PSD-95 m RNA: The expression of PSD-95 m RNA in the model group first decreased(7-14d)and then increased(21-28d),and the expression decreased to the lowest on 14 d.Pericardial meridian group:14d>7d>3d(p<0.01,p<0.05)),28d>21d(p<0.01)and basically the 3d(p<0.01).and the pericardial meridian group was better than the lung meridian group.Conclusion:1.Electroacupuncture at the pericardium meridian and lung meridian points can promote the expression of SYP,PSD-95 and m RNA in MCAO model rats and cerebral infarction area,and the pericardium meridian group is better than the lung meridian group.Acupuncture can affect cerebral infarction as time goes by The role of PSD-95 in the area gradually increased.2.Electroacupuncture at the points of the pericardium meridian may participate in the regulation of synaptic plasticity by promoting the expression of SYP/PSD-95 in the cerebral infarct area of MCAO rats.
