Objective:To explore Professor Wang Xing kuan’s medication experience in the treatment of hypertension and extract the core prescription by data mining;At the same time,a network pharmacological analysis of the core prescription for the treatment of hypertension was carried out to explore its mechanism of action.Through the above research,Professor Wang Xing kuan’s clinical experience in treating hypertension is summarized in order to guide the clinic.Methods:1.Data mining research: Collect the effective medical records of Professor Wang Xing kuan’s outpatient treatment of hypertensive patients at the outpatient from January 2018 to December 2020 to establish a database,and enter the data into the Traditional Chinese Medicine Inheritance Computing System(V3.0),based on the platform Provide a variety of data mining methods,combined with Professor Wang Xing kuan ’s typical medical records for text study,analyze the basic properties of commonly used Chinese medicines and drugs in the prescriptions of medical records,explore drug combinations,refine core prescriptions,and summarize Professor Wang Xing kuan ’s prescriptions for the treatment of hypertension.2.Net wor k pharm ac ol o gy res e arc h: us e TCM S P and BATM A NTCM databases to collect the core prescription active ingredients and corres ponding targets,obtain hypertension disease targets th r ough Gene Cards 、 Dis Ge NET 、 OMIM 、 and TTD databases,and bui ld th e core with Cytoscape 3.8 so ftware The "active ingredient-d iseas e target" netwo rk diagram of the prescription for the treatm en t of hypertension;the potential target interaction network diag ram is est ablished based on the STRING database,an d the key ta rgets are screened according to the degree value parameters;final ly,the Met as cape da tabas e is us ed to enric h the pot enti al targets with GO and KEGG.Set an alys is to explore the mechanis m of a ction of core prescriptions in the treatment of hypertension.Results:1.Data mining research : A total of 354 prescriptions involving134 Chinese materia medicines were included.The top 10 Chinese medicines used most frequently are Rhizoma Gastrodiae,Ramulus Uncariae Cum Uncis,Terrestris Tribulus Fruit,Rhizoma Dioscoreae,Fructus Corni,Radix Rehmanniae Preparata,Rhizoma Alismatis,Concha Haliotidis,Radix Puerariae,Poria Cum Radix Pini,etc.;The proportions of four nature of drugs used are calm,cold,warm,cool,and hot in order from high to low;The proportions of the five flavors of drugs used are sweet,bitter,pungent,sour,and salty from high to low;The proportion of the channel entry from high to low is liver,kidney,spleen,heart,lung,stomach,gallbladder,pericardium,bladder,large intestine,small intestine,and sanjiao meridian;The top 5 in order of the proportion of efficacy are in order to invigorate deficiency,calming the liver to stop the wind,increasing secretion of urine,promoting blood circulation for removing blood stasis,and clearing heat.The number of support degrees set based on the association rule is 177.The confidence level was 0.9,and the group of 214 were counted as the more closely related common drug combinations,of which the frequency of the hemp-hook rattan drug pair was the highest,343 times;The core prescription components include Rhizoma Gastrodiae,Ramulus Uncariae Cum Uncis,Terrestris Tribulus Fruit,Concha Haliotidis,Rhizoma Dioscoreae,Fructus Corni,Radix Rehmanniae Preparata,Rhizoma Alismatis,Radix Puerariae 9 flavor Chinese materia medicine;Based on k mean clustering,6 optimal clusters were obtained based on the combination of prescription-like drugs except core prescription and core drug law.2.Network pharmacology research: After screening,there are 64 active ingredients in the core prescription,including quercetin,kaempferol,stigmasterol,and beta-sitosterol,which cross-act 67 hypertension targets,and the key targets include IL6,FOS,TNF,NOS3,VEGFA,etc.;GO biological function enrichment yields 79entries;KEGG enrichment analysis yields a total of 84 pathways,Among them,it is mainly involved in multiple signaling pathways including MAPK,HIF-1,P13K-Akt,Calcium signaling pathway,Fluid shear stress and atherosclerosis,and Vascular smooth muscle contraction.Conclusion:1.Professor Wang Xing kuan’s treatment of hypertension follows the basic principles of regulating liver and kidney yin and yang.The treatment method is mainly to invigorate the kidney and calm the liver,take care of dredging the blood,and the prescription is flexible.The same function,supplementing and reducing both,embodies Professor Wang’s academic thought of "regulating multiple internal organs,treating miscellaneous diseases and treating liver".2.The core prescription works by multi-components,multi-targets and multi-channels at the same time,and predicts that it will mainly mediate inflammation and oxidative stress,improve vascular endothelial function,and play a comprehensive regulatory role on blood pressure.
