Purpose:Based on data mining technology,this paper explores the prescription rule of Professor Wang Fengrong in treating hypertension,and explores the potential mechanism of his core prescription in treating hypertension by means of network pharmacology research method,.providing reference for TCM treatment of hypertension.Material and methods: From May 2018 to August 2020,the clinical data of patients with hypertension treated in Professor Wang Fengrong’s outpatient clinic of the Affiliated Hospital of Liaoning University of Traditional Chinese Medicine were collected.According to the inclusion criteria,78 patients were included and 167 prescriptions were entered.Excel was used to establish the patient information database,analyze the general situation of patients and the distribution of syndromes,and count the drug frequency.SPSS Modeler 18.0and IBM SPSS Statistics 19 software were used to analyze the association rules and cluster analysis of prescription information,and the core prescriptions were selected based on the above analysis results The chemical components of the core prescription were obtained with the help of TCMSP and BATMAN-TCM databases,which were supplemented by the literature mining of CNKI.With the oral bioavailability(OB)≥30% and drug-like property(DL)≥0.18 as screening conditions,the effective components and target information of the core prescription were obtained from TCMSP database,and the target was converted into corresponding gene names through Uniprot database.The gene target of hypertension was obtained by using the Dis Ge NET database,the network of "drug-component-target-disease" was constructed by using the software of Cytoscape3.7.2,the PPI network was constructed by using the STRING database,and the pathway analysis was carried out by using David and KEGG database,so as to predict the action mechanism of the core prescription in treating hypertension.Results:1.Phlegm dampness(48 cases)and yin deficiency(44 cases)are the most common syndromes in patients with hypertension treated by Professor Wang Fengrong.The five most frequently used drugs are Glycyrrhiza uralensis Fisch(5.17%),Uncaria rhynchophylla(4.63%),Gastrodia elata Blume(4.56%),Achyranthes bidentata(3.91%)and Pueraria lobata(3.87%).The drug pairs with high support in association rule analysis are Gastrodia elata and Uncaria,Gastrodia elata and Achyranthes,Uncaria and Achyranthes,Gastrodia elata and Pueraria lobata,Uncaria and Pueraria lobata.Drug combinations with 100% confidence include Gastrodia elata-Uncaria-Achyranthes,Gastrodia elata-Uncaria-Achyranthes-Pueraria lobata,Gastrodia elata-Uncaria-Achyranthes-Glycyrrhiza uralensis Fisch.The core compound is Gastrodia elata Blume-Uncaria rhynchophylla-Achyranthes bidentata-Pueraria lobata.2.The results of cluster analysis showed that the drugs used frequently ≥55 could be clustered into three groups,including keel-oyster-arborvitae seed,Uncaria rhynchophylla-Gastrodia elata-Achyranthes root-kudzuvine root-chrysanthemum-concha Haliotidis-Spica Prunellae-Scutellaria baicalensis Georgi-motherwort,and Acorus calamus-Radix Curcumae-Polygalae.3.The "drug-component-target-disease" network cross-mapping shows that the higher degree value components of the core compound are quercetin,kaempferol,wogonin,baicalein,rhynchophylline,phytosterol and conaine.It is predicted that these components are mainly controlled by PTGS2,HSP90AB1,PTGS1,AR,SCN5 A,F10,KCNH2,CHRM1,CHRM3 and ADRA1 B.4.GO and KEGG enrichment analysis core compound mainly participates in biological processes such as autophagy,apoptosis,proliferation and differentiation through PI3K-Akt signal pathway,MAPK signal pathway,cell aging pathway and Ras signal pathway.Conclusion:1.Professor Wang Fengrong focuses on liver and spleen in TCM treatment of outpatients with hypertension,and the core compound for treating hypertension is "Gastrodia elata-Uncaria-Achyranthes bidentata-Pueraria lobata".2.Based on the research of network pharmacology,"Gastrodia elata-Uncaria-Achyranthes bidentata-Pueraria lobata" drug combination contains 49 effective chemical components,among which quercetin,kaempferol,wogonin,baicalein,β-sitosterol,yohimbine,rhynchophylline,phytosterol and conaine have anti-inflammatory,anti-tumor,anti-oxidation,and can improve ventricular remodeling and vascular endothelial damage.3.Based on network pharmacology,it is predicted that "Gastrodia elata-Uncaria rhynchophylla-Radix Puerariae" mainly regulates PTGS2,HSP90AB1,PTGS1,AR,SCN5 A,F10,KCNH2,CHRM1,CHRM3,ADRA1 B,etc.,and participates in the regulation of PI3K-Akt signal pathway,MAPK signal pathway,cell aging pathway and Ras signal pathway.
