| Objective: To observe the effect of Zhixiang tablets on the changes of SOD,MDA and ultrastructure of skeletal muscle after exercise-induced muscle injury in SD rats,and analyze its protective mechanism on exercise-induced muscle injury,so as to provide theoretical basis for the prevention and treatment of exercise-induced skeletal muscle injury.Methods: One hundred and five SD rats were randomly divided into group A0(quiet control group),group A(exercise only group)group B(Zhixiang tablets group),and group C(soy peptide group),with no intervention in group A and fragrance tablet and soy peptide in groups B and C,respectively,one week before modeling.The animal model of skeletal muscle injury caused by a single large-load centrifugal exercise was established by performing 5 min×18 times at 26.8 m/min on a-16°running table for a total of 90 min,with a 1 min interval between groups.The content of propylene glycol(MDA)and superoxide dismutase(SOD)in quadriceps muscle homogenate was measured at five time points immediately after exercise,12 h,24h,48 h and 72 h,and the ultrastructural changes of quadriceps muscle were observed with the help of transmission electron microscopy.Results:(1)after a heavy load eccentric exercise,the content of MDA in groups A,B and C reached the peak immediately after exercise,and then decreased gradually.The content of MDA in group B was significantly different from that in AO group at immediately,12 h and 24 h after exercise,but there was no significant difference between group B and group AO at 48 h and 72 h after exercise.There was a significant difference in the content of MDA between group B and group An at the same time after exercise(P < 0.05).There was no significant difference between group B and group C after exercise(P > 0.05).(2)After a heavy load eccentric exercise,the content of SOD in groups A,B and C suddenly decreased to the lowest immediately after exercise,then gradually increased at different time points and reached the peak at 48 h.There was no significant difference in the content of SOD between group B and group An immediately and 12 hours after exercise,but there was significant difference between group B and group An at 24 h,48 h,72 h after exercise.The content of SOD in group B immediately after exercise was significantly different from that at24 h,48 h and 72 h after exercise(P < 0.05).There was no significant difference between group B and group C after exercise(P > 0.05).(3)From the histomorphological observation,the 24 h electron microscopy of the simple exercise group showed disruption of muscle structure,disorganized arrangement of myogenic fibers,sawtooth-like changes in Z-lines,blurred demarcation of I-band and A-band,and swelling of some mitochondria.In the aroma tablet and soy peptide groups,the myofibrils were more neatly arranged and the boundaries of the I and A bands were clearer than in the soy peptide group.The myofibrillar arrangement and the boundaries of the bands were clearer in the aroma tablet group compared with the soy peptide group.Conclusion:The Zhixiang tablets has good effect on preventing the exercise-induced soft tissue injury of rats.Its mechanism may be through reducing the content of MDA in muscle,improve the activity of SOD,improve the antioxidant capacity of muscle,remove excess free radicals,so as to effectively protect the cell membrane and cell structure and function. |
