Objective: To investigate the changes of CK-MM,s Tn I and skeletal muscle ultrastructure of Chinese patent medicine Zhixiang tablets on exercise-induced muscle injury induced by a high intensity eccentric exercise.Method: 99 SD rats were randomly assigned to group A(exercise group),group B(soybean peptide group),group C(Zhixiang tablets group).Group A simply complete the eccentric exercise.Group B and group C take the soybean peptide and the Zhixiang tablets respectively a week before modling,and stopped taking the medications the day the model was made.A week later all the rats on the-16°platform running at a speed of 26.8 m/min to 5 min×18 times 90 minutes in all of high intensity intermittent treadmill exercise,reseting for 1 min each 5 minutes.The rats in each group were sacrificed at Oh,12 h,24h,48 h and 72 h respectively.The muscle homogenates of the rats were collected to measure the contents of CK-MM and s Tn I.The histomorphological changes of the three groups under high resolution electron microscope were observed at 24 h and 72 h respectively.Results: s Tn I of group B and group C during 0h--12 h was significantly lower than that of group A(p<0.05),and the concentration of each group was similar at 24 h without significant difference(p>0.05).The increase of s Tn I concentration during48h--72 h was higher than that of group A,and there was a significant difference between group A and group C(p<0.05).The CK-MM of group B and group C at 0h was significantly higher than that of group A(p<0.05),and decreased rapidly at 12 h,but there was no significant difference among the three groups(p>0.05).At 24 h,the CK-MM of group C and group B was slightly increased,then decreased rapidly but there was no significant difference among the three groups(p>0.05).In the early stage of injury(0h-12h),the release of CK-MM and s Tn I was rapidly reduced by both the Zhixiang tablets and the soybean peptide,but the therapeutic effect was not obvious in the subsequent period,and there was no significant difference in the curative effect between the Zhixiang tablets and the soybean peptide.Electron microscope sections showed that at 24 h,the myofibrillars in the group A were arranged disorderly,adjacent muscle segments were distorted,Z-lines and M-lines were obviously distorted and deformed,and “serrated” changes appeared,and the muscle space became wider.The myofibrils of the group B were clearly arranged,the bright and dark bands were not clearly identifiable,and the Z-lines and M-lines were distorted and deformed.In the group C,the myofibrils were clearly arranged,the bright and dark bands were clearly identified,the Z-lines and M-lines were less distorted and deformed,and some muscle segments were slightly deformed.At 72 h,the injury was relieved in the group A compared with 24 h,Z-lines and M-lines were arranged more neatly,and some Z-lines were lost and blurred.The injured myofibrils began to hydrolyze and blur,and a large number of long mitochondria were seen.In the group B,the myofibrils were hydrolyzed and blurred,and the mitochondria were disordered.There were long strips and swollen mitochondria in different shapes and sizes.In the group C,the morphology of Z-lines,M-lines,light and dark bands and sarcomaglia were clearly discemible,and the myofilaments were clear without continuity or large area blurring.The morphology of mitochondria was normal and the distribution was symmetrical.Conclusion: 1.The Zhixiang tablets can effectively inhibit the release of CK-MM and s Tn I in skeletal muscle at the early stage of injury(0h-12h),and protect the integrity of cell membrane.2.the Zhixiang tablets have been observed to significantly reduce the ultrastructural injury,suggesting that the Zhixiang tablets can prevent the eccentric exercise induced-muscle damage.
