Purpose:The expression of 4-HNE in oxidative stress was studied,and the mechanism of Huatan Sanjie granule in the treatment of iodine deficiency goiter was studied.Material and method::A total of 1824-week-old Wistar rats,half male and half female,were randomly selected and fed with conventional diet.After one week,30 rats were randomly selected as normal control group(NC)and fed with conventional diet.The other 152 rats were fed with low iodine diet and sodium perchlorate for 19 days,followed by double distilled water for 2 days.After 21 days,two rats were randomly selected to verify the goiter model.After successful modeling,they were randomly divided into 5 groups with 30 rats in each group as: model control group(MC),excess iodine group(PHI),levothyroxine sodium tablet group(L-T4),Huatan Sanjie granule group(Hg)and Huatan Sanjie Granule plus levothyroxine sodium tablet group(Hg + L-T4).MC was given equal volume of deionized water;Phi was given equal volume of high iodine water containing iodine 1900 μ g / L;Hg was given equal volume of Huatan Sanjie granule concentrated decoction converted by body surface area,Each rat was given 20.52 g / kg of crude drug every day;L-T4 was given the same volume of levothyroxine sodium tablets solution;Hg + L-T4 was given the same volume of Huatan Sanjie granule concentrated decoction and levothyroxine sodium tablets solution;once a day,a total of 77 days.The rats were killed 21 days and 77 days after the intervention,15 rats in each group.Abdominal aorta blood collection and thyroid tissue separation were performed under abdominal anesthesia.Thyroid tissue morphology was observed by HE staining.TT3 and TT4 in serum were measured by RIA.TSH in serum was measured by IRMA.Thyroid index was calculated by dividing wet weight of thyroid by body weight.The expression of 4-HNE was detected by immunofluorescence.Results:1.Model replication: after 21 days of gavage with low iodine diet,sodium perchlorate and double distilled water,it can be seen by light microscope,Compared with NC,MC thyroid follicles were significantly different in size,small and irregular in shape,with less colloid in the cavity.Some follicles fused with each other to form giant follicles.There were a lot of inflammatory cells infiltration and follicular hyperplasia in the follicular cavity and stroma.There were abundant blood vessels around the follicles,which suggested that the rat model of goiter was successfully established.2.HE staining of thyroid cells: compared with NC,MC had inflammatory infiltration in the thyroid follicles,the follicular cavity disappeared,the blood vessels around the follicles were abundant,the morphology of epithelial cells changed,and the double membrane structure disappeared.Compared with Phi group,the epithelial cells of thyroid follicles increased in Hg and L-T4 groups.The follicular epithelium in Hg + L-T4 group was flat and high,which was higher than that in L-T4 group and Hg group.3.Thyroid function of rats in each group: compared with NC,TT3 and TT4 in other groups were significantly decreased and TSH was significantly increased on the 21 st day.On the77 th day,compared with NC,TT3 in Hg group and L-T4 group were significantly increased,and TSH values in other groups were still significantly increased.On the 21 st day,compared with MC,TT3 and TT4 in other groups were significantly increased,while TSH was significantly decreased.On the 77 th day,compared with MC,TT3 and TSH in p Hi group and Hg group were significantly lower,while TT4 was significantly higher.4.Comparison of relative mass of thyroid: compared with NC,the relative mass of thyroid in other groups increased significantly on the 21 st day(P < 0.05).After 77 days,compared with MC,The relative quality of thyroid in other treatment groups decreased significantly(P<0.05).The relative mass of thyroid in Hg group,Hg + L-T4 group and L-T4 group decreased,but the difference was not statistically significant(P > 0.05).5.Expression of 4-HNE in thyroid: compared with NC,the inflammatory exudation of thyroid cells in other groups increased.Compared with MC,the inflammatory exudation of thyroid cells in L-T4 group,Hg group and Hg + L-T4 group decreased significantly.Compared with L-T4 group,the inflammatory substances in Hg + L-T4 group were significantly reduced.Conclusion:1.the combination of traditional Chinese and Western medicine,Huatan Sanjie granule and L-thyroxine sodium tablet,can better inhibit the proliferation of thyroid cells,make the thyroid cells not damage the original thyroid cells while recovering,and play a therapeutic role in the treatment of thyroid cancer by regulating the expression of 4-HNE in the oxidative stress response of goiter.
