Purpose: By analyzing the effective chemical components of Shugan Huazhi Capsules and studying its mechanism of action in the treatment of non-alcoholic fatty liver disease(NAFLD),it provides a solid theoretical basis for the clinical application of Shugan Huazhi Capsules in the treatment of NAFLD.Methods: Through TCMSP database,ETCM database,Pub Chem database,SIB database and other databases to query the effective targets of drugs,the seven traditional Chinese medicines of Shugan Huazhi Capsules(Salvia,Bupleurum,Pinellia,Cassia,Turmeric,Alisma,Hawthorn))Effective chemical active ingredients and target proteins corresponding to the chemical ingredients and acting on the human body to search,screen,summarize and summarize.Use the OMIM database and Gene Cards database to predict the target of NAFLD disease,integrate the effective chemical components of NAFLD from the two databases,and finally query the protein names and corresponding gene names of the corrected drugs and diseases through the Uni Prot database,and perform the two through the Excel table Integrate and screen the duplicate targets of the two.Use Cytoscape software to construct a drug-diseaseactive ingredient target network diagram to achieve visualization;construct a protein interaction network through the String database platform;obtain GO(gene ontology)enrichment analysis and KEGG(Kyoto Encyclopedia of Genes and Genomes)through DAVID database)Enrichment analysis;use the Omic Share Tool cloud platform to visualize the results and construct a target-path network diagram.Confirmatory experiment: 30 healthy SD rats are guaranteed to be male and have normal diet.30 rats are randomly divided into three groups,10 normal group,10 model group,and 10 traditional Chinese medicine group.The normal group was fed with conventional feed for 8 weeks,followed by normal saline gavage for 4 weeks;the model group was fed with high-fat feed for 8 weeks,followed by normal saline gavage for 4weeks;the traditional Chinese medicine group was fed with high-fat feed for 8 weeks,After that,a traditional Chinese medicine gavage(Shugan Huazhi Capsule Powder)was given for 4weeks.Observe the levels of interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α)in serum samples by enzyme-linked immunosorbent assay(ELISA);real-time fluorescent quantitative PCR(Quantitative Real-time PCR)to detect the relative expression of Toll like receptor-4(TLR-4)mRNA and nuclear factor-κB(NF-κB)mRNA in liver tissue.Result:1.After screening,70 effective ingredients of Danshen,22 effective ingredients of Pinellia ternata,29 effective ingredients of Bupleurum,24 effective ingredients of cassia seed,6effective ingredients of turmeric,and effective ingredients of Alisma There are 14 ingredients and 21 active ingredients in hawthorn.2.There are 1029 NAFLD disease targets,and a total of 168 drug-disease common target genes have been obtained.3.Through the PPI protein interaction relationship,the 27 core genes are INS,ALB,AKT1,IL6,TNF,TP53,VEGFA,JUN,MAPK8,CASP3,STAT3,EGF,EGFR,PTGS2,MYC,CAT,MMP9,PPARG,ESR1,IL1 B,CXCL8,TLR-4,CYCS,ACTB,CCL2,NOS3,CCND1;4.GO enrichment analysis results include cell response to lipopolysaccharide,inflammation,exosomes,membrane rafts,enzyme binding,protein binding,etc.;5.KEGG enrichment pathway includes NF-κB signaling pathway,tumor necrosis factor signaling pathway,AMPK signaling pathway,etc.6.Confirmatory research results: Compared with the normal group,the serum IL-6 and TNF-αlevels and the liver tissue TLR-4 mRNA and NF-κB mRNA expression levels in the model group were significantly increased(P<0.05);and Compared with the model group,the serum IL-6 and TNF-α levels and the liver tissue TLR-4 mRNA and NF-κB mRNA expression levels in the traditional Chinese medicine group were significantly reduced,and the difference was statistically significant(P<0.05).Conclusion:1.The key chemical components of Shugan Huazhi capsule in the treatment of NAFLD are quercetin,luteolin,kaempferol,β-sitosterol,tanshinone IIA,baicalein,etc.,which have a high expression effect.2.Shugan Huazhi Capsule may reduce inflammation by regulating IL6,TNF,TLR-4 and other targets,or regulate NF-κB signaling pathway to affect inflammation,and then play a role in the treatment of NAFLD.
