Effect Of ShuganXiaozhi Recipe Ⅲ Capsule On The CYP2E1 In The Rat With Non-alcoholic Fatty Liver Disease

Objectives:To study the therapeutic effects of Shugan Xiaozhi Recipe III capsule on the expression of hepatic cytochrome P4502E1 m RNA in the rats with non-alcoholic fatty liver disease(NAFLD),and try to make further analyses of its action mechanism.And to provide scientific experimental basis for clinical trials and safe medication.Methods:Fourty-six healthy male SD grade specific pathogen-free(SPF)rats were randomly divided into four groups,which included a normal diet control group(n=10)and the NAFLD model group(n=12),Silibnin capsules group(n=12)and SXRC Ⅲcapsule group(n=12).The normal group had a regular diet and were given a gavage of physiological saline of the same volume;the NAFLD model group and the drug group were given a gavage of high-fat emulsion according to the 1 ml / 100 g weight.After six weeks the orbital blood was collected for measuring the levels of the triglyceride(TG),the total cholesterol(TC),and extracted 2 rats randomly from model group and the drug group for pathological observation which suggests the success of the model.From the seventh week,the experimental rats of Silibnin capsules group and SXRC Ⅲcapsule group were given drug by intragastric administration.The normal group and the NAFLD model were daily given physiological saline(1 ml / 100 g);Drug groups were respectively given silibinin capsules(44.1 mg/kg)and SXRC Ⅲcapsule(315 mg/kg)by intragastric administration every day.After four weeks of treatment(by the end of the 10 th week),the mental state and surface gloss,activity of the rats were observed.Their body length and weight were measured,and then all rats were killed by the method of cervical dislocation.Their blood was collected and acupuncture serum(AS)was prepared.After harvesting the liver,the whole liver wet weight were weighted and the liver index was counted.After making conventional preparation of liver homogenate and liver tissue paraffin section of the specimen,the left liver tissue were put in – 80℃ refrigerator,which were for q PCR detection.The content of each group of serum aspartate aminotransferase(AST),alanine aminotransferase(ALT),cholesterol(TC),triglyceride(TG),low density lipoprotein(LDL-C),high density lipoprotein(HDL-C)were detected and analysised.The CAT activity in liver homogenate were detected by the method of visible light.The level of liver tissue CYP2E1 m RNA was detected by the method of Fluorescence quantitative(QPCR),and HE staining method was used to to observe the liver tissue pathological changes of each group.Results:1.Compared with the control group: in NAFLD group the levels of liver index and the contents of serum biochemical indexes(ALT,AST,TC,TG,HDL-c,LDL-c),and CYP2E1 m RNA of the rats were all elevated(P<0.05),the body weight and body length of the rats growth more slowly(P<0.01),the CAT activity had decreased in liver homogenate(P<0.01)and Liver Histopathology showed moderate to severe steatosis.2.Compared with the control group: in SXRC Ⅲcapsule the levels of liver index and the contents of serum biochemical indexes(ALT,AST,TC,TG,LDL-c),and CYP2E1 m RNA had decreased(P<0.05),but the body weight and body length had increased(P<0.05),the CAT activity had increased in liver homogenate(P<0.01)and fatty degeneration of the liver tissue and inflammation had lessened.3.Compared with silibinin capsules group: in SXRC Ⅲcapsule the levels of liver index and the contents of serum biochemical indexes(ALT,AST,TC,TG,HDL-c,LDL-c),and CYP2E1 m RNA had reduced,but the differences made no sense(P>0.05),the body weight and body length of the rats had slightly increased,but the differences of statistics made no sense(p>0.05);the CAT activity had increased in liver homogenate(P>0.05);the difference of the liver tissue pathological changes were resemblely.Conclusion:Shugan xiaozhi capsule III was a new treatment method of NAFLD,because it can improve metabolic disorders,liver function and liver steatosis and hepatic inflammation of the NAFLD rats.High-fat diet can lead to the decrease of antioxidant CAT activity and the expression of CYP2 Elm RNA was significantly higher in NAFLD rats,Shugan xiaozhi capsule III can significantly increase the activity of CAT and inhibit the expression of hepatic CYP2E1 gene in cells,thus reducing oxidative stress and lipid peroxidation.
