Objective: To observe the effect of Jiedu Huayu Shengjin Decoction on general condition,pathological changes of submandibular gland and expression of related cytokines in serum of Sjogren’s syndrome model mice,and to explore its mechanism of action.Materials and methods: 60 C57BL/6 mice were randomly divided into normal group,model group,hydroxychloroquine sulfate group,Jiedu Huayu Shengjin prescription low-dose group,medium-dose group and high-dose group.Submandibular gland immune induction method is adopted to establish the sjogren’s syndrome animal model of mice with detoxification blood party to lavage treatment of experimental mice,with Western medicine Hydroxychloroquine Sulfate tablets(Hydroxychloroquine Sulfate,HCQ)lavage in comparison,observation groups of mice with and without hair removal,surface gloss,weight,water content,food intake,saliva secretion,submandibular gland index,using ELISA method and Western Blot method to detect the mice IL-17,IL-4,IL-6,the expression of TNF alpha,HE staining observation of submandibular gland pathological changes.Results:1.General situation of mice: Jiedu Huayu Shengjin Formula can effectively improve the general states of SS model mice,increase body weight and food intake,significantly relieve the state of dry mouth,increase the amount of saliva,and reduce the amount of drinking water;The index of submandibular gland and thymus of mice in the model group decreased significantly,and the index of submandibular gland and thymus of mice in each administration group was between the model group and the normal group.Jiedu Huayu Shengjin formula significantly reduced the atrophy of submandibular gland and increased the index of submandibular gland,and the effect was more obvious in the high-dose group.2.Mouse submandibular gland tissues HE staining results,compared with normal group,model group mice submandibular gland cells clearance increases,glands severe damage of substance within the stroma is easy to see multiple focal lymphocytic infiltrates,partial gland shrink,fester,acini damage,incomplete,and so on and so forth,visible gland reactivity control cavity expansion at the same time,the blood vessel wall thickening,building success.In the low-dose group,the space between submandibular gland cells increased unevenly,the glands were moderately damaged in parenchyma,1-2 lymphocytes were occasionally observed in the stroma,and the acinus was damaged,incomplete,and of different sizes.The gland lumen was reactive-dilated,the blood vessel wall was thickened,and some capillaries were congested.In the medium dose group,the cell space of submandibular gland was enlarged,and lymphocytes were infiltrated in the interstitium of the gland with mild parenchymal injury,but no lymphatic foci were observed.In the high-dose group,the space of submandibular gland cells increased,lymphocyte infiltration was rare,acinus damage was not obvious,and the size was close to the same,the gland lumen expanded and atrophy,blood vessel wall thickened,and capillary congested.In the hydroxychloroquine group,the cell space of the submandibular gland was enlarged,and a small amount of lymphocytes were infiltrated in the intercellular stroma,and the acinar damage was not obvious,and the size was close to the same.The glandular lumen was dilated and atrophied,and the vascular wall was thickened.3.The contents of IL-4,IL-6,IL-17 and TNF-α in mice serum were determined by ELISA method.The results showed that there were low concentrations of IL-4,IL-6,IL-17 and TNF-α in serum of normal mice.The concentrations of IL-4,IL-6,IL-17 and TNF-α in serum of mice in the model group were the highest,and the concentrations of IL-4,IL-6,IL-17 and TNF-α in serum of mice in the high-dose Jiedu Huayu Shengjin prescription group were the most close to the normal group.4.The expressions of IL-4,IL-6,IL-17 and TNF-α in the submandibular gland of mice were detected by Western Blot.Compared with the normal group,the expressions of IL-4,IL-6,IL-17 and TNF-α in the high-dose group and the hydroxychloroquine group were not significantly different,while the expressions of IL-4,IL-6,IL-17 and TNF-α in the other groups were significantly increased,and the differences were significant.(2)Compared with model group,the expressions of IL-4,IL-6,IL-17 and TNF-α were significantly decreased in all groups except the low-dose group;(3)Compared with the hydroxychloroquine group,the expression of IL-4,IL-6,IL-17 and TNF-α in the high-dose group was not significantly different,while the expression of IL-4,IL-6,IL-17 and TNF-α in the other groups was significantly higher.Conclusion:1.Both Jiedu Huayu Shengjin Formula and Hydroxychloroquine can significantly improve the symptoms of SS.2.The traditional Chinese medicine Jiedu Huayu Shengjin Formula can reduce inflammation by down-regulating IL-4,IL-6,IL-17 and TNF-α.3.The prescription of Chinese medicine for detoxification,removing stasis and Shengjin can reduce the pathological injury of the submandibular gland,and the high and medium doses of Chinese medicine have the best efficacy in the Chinese medicine group.
