Hypertension is an important risk factor for cardiovascular disease,and its incidence is increasing year by year.The renin-angiotensin-aldosterone system plays a crucial role in the regulation of blood pressure.Its main function is to regulate and maintain the balance of blood pressure,water and electrolytes in the human body,and maintain the relative stability of the internal environment.Excessive activation of renin-angiotensin-aldosterone system is one of the causes of high blood pressure.In this study,the correlation activities of four common Chinese olives on the market were measured;the effects of different extracted solvents were studied,and correlation analysis was carried out to explore the connection between the main chemical components and activities,and to speculate on the types of substances that play a dual inhibitory role;the single component with the dual inhibition effect of renin and angiotensin converting enzyme(ACE)was obtained by separation and purification,the enzyme inhibition type was determined,and the relevant data of the structure were obtained by liquid-mass spectrometry.To provide basic data and theoretical basis for the development of olives and the study of foodborne renin and ACE inhibitors.Different varieties of Chinese olives showed certain renin and ACE inhibitory activities.Under the condition of 0.5μg/μL concentration,the inhibition rate in renin suppression experiments reached more than 70%,of which the renin inhibition rate of Huiyuan was the highest,at 83.93%.Under the condition of 0.29μg/μL concentration,the inhibition rate of ACE inhibition in the experiment reached more than 80%.Among them,Huiyuan had the highest ACE inhibition rate,which was 94.75%.In the antioxidant experiment,the antioxidant capacity of Tanxiang was the best among the four types of olives.The free radical scavenging capacity of ABTS,DPPH radical scavenging capacity,and iron ion reduction capacity reached 182.64 mg TE/g DW and 140.64 mg TE/g DW and 251.65 mg TE/g DW,respectively.The antioxidant capacity of Sanleng is second only to that of Tanxiang.Overall,olive extracts showed a certain strength of antioxidant capacity.Tanxiang has the highest total phenol content at 274.56 mg GAE/g DW.In terms of total flavonoids,Wulan has the highest content at 52.64 mg CE/g DW.Chinese olives were extracted from 70%methanol,60%ethanol,60%acetone,and pure water.The renin and ACE inhibitory activities of the organic solvent extract were generally higher at the same concentration,and the renin inhibitory activity of methanol extract reached the highest 95.89%,and the ACE inhibitory activity of acetone extract reached the highest92.71%.Similar to renin and ACE inhibition,antioxidants were also more capable of organic solvent extracts.In terms of ABTS free radical scavenging,acetone extract reached 384.86mg TE/g DW.In terms of DPPH free radical scavenging,it is also acetone extract.It had the strongest ability,reaching 309.23 mg TE/g DW,and the iron ion reducing ability of acetone extract is 479.40 mg TE/g DW.Acetone extract had the highest total phenol and total flavonoid content at 318.14 mg GAE/g DW and 45.46 mg CE/g DW.Pearson correlation analysis showed a significant correlation between renin and ACE inhibition and total phenol content,and it was speculated that the substances that played a role in renin and ACE inhibition were phenolic.The macroporous adsorption resin chromatography and high performance liquid chromatography were used to separate and purify the renin and ACE inhibitory active components in the olive extract of Tanxiang,and obtain the single components F1,F2,F3.The molecular weight of component F1 is 269.11 m/z,and the IC50 values of renin and ACE inhibition are 161.08 and 48.85μM,respectively;the molecular weight of component F2 is206.10 m/z,and the IC50 value of ACE inhibition is 174.28μM;component F3 Its molecular weight is 570.19 m/z,and its IC50 values for renin and ACE inhibition are 9.63 and 64.39μM,respectively.Combined with literature reports,it is inferred that component F2 is 6,7-dimethoxycoumarin.Fractions F1 and F3 showed dual inhibition of renin and ACE,and the inhibition types of renin and ACE were both anti-competitive inhibition,which provided an important basis for in-depth study of blood pressure regulators in olives. |
