Aging is a common physiological process of all living things,and its internal mechanism and causes are extremely complex.In the face of the increasingly severe trend of population aging,the extension of human life expectancy also means the increase of the probability of aging and its related diseases.Therefore,how to reduce the harm of aging and its related diseases to human health has gradually become one of the hot issues in recent years.The application of natural plant molecules in anti-aging,stress response,neuroregulation and other research fields has attracted attention due to their advantages of wide sources,small toxic and side effects,and low difficulty in structural analysis.Functional foods with natural plant monomers as the main ingredients are also well known by the public.Therefore,the research and development of natural plant monomer function,in addition to the in-depth exploration of natural plant function,is also of great significance in the development of its application in functional food.Previous studies have shown that a large number of natural plant antioxidants have anti-aging effects.Ellagic acid is a kind of polyphenolic monomer compound that exists in strawberries,pomegranates and other fruits,and has strong free radical scavenging ability.However,the role of ellagic acid in anti-aging is still not well studied.In this work,the anti-aging effect of ellagic acid on C.elegans and its related mechanism were studied.The results showed that compared with the control group,ellagic acid significantly prolonged the lifespan of C.elegans by 24.06%,and the effect of prolonging the lifespan was not through affecting the growth of E.coli OP50;On this basis,ellagic acid has no harmful effect on the growth and reproduction of C.elegans,while improving its locomotion ability and reducing its lipofuscin accumulation;In the stress environment,ellagic acid can also improve the survival rate of C.elegans under emergency stress,and reduce the ROS accumulation level caused by stress,in which the ROS level is reduced by about 36.9% under heat stress and about 57.3% under paraquat stress.Using the lifespan measurement of the mutants of C.elegans,it was found that the effect of ellagic acid on prolonging lifespan disappeared in the mutants of daf-2,daf-16,skn-1,hsf-1 and hlh-30,Indicating that the anti-aging function of ellagic acid is achieved through the insulin/IGF-1 signaling pathway;The results of q RT-PCR further showed that the m RNA expression levels of the corresponding genes and their downstream target genes of the above mutant nematodes treated with ellagic acid were also up-regulated.The effect of ellagic acid on the gene changes of C.elegans was investigated by transcriptomic sequencing.KEGG and GO analysis showed that ellagic acid enriched different genes in a large number of metabolic pathways of C.elegans;KEGG analysis showed that in addition to the longevity regulation pathway,ellagic acid also showed significant regulatory activity in the MAPK signal pathway of C.elegans.In conclusion,this work proves that ellagic acid has strong anti-aging and anti-stress activation,preliminarily clarifies the mechanism of ellagic acid delaying the aging of C.elegans through insulin/IGF-1 signal pathway,and can activate the expression of its downstream antioxidant gene and detoxification gene to inhibit the accumulation of ROS in C.elegans.The above research results lay a foundation for the application of ellagic acid in anti-aging and other fields. |
