At present,the studies on the chemical compounds of Lilium mainly focus on the bulb,while there are few reports on other parts.There are many kinds of lilies.Some lilies have bitter taste,while others have no.What is the bitter substance in lilies,there has been no report.From 1985 to 2020,there are many studies reported that colchicine was contained in lily,and our previous studies preliminarily found that colchicine may not exist in lily.Whether colchicine exists in lily is a controversial issue.In view of the above three main scientific problems,this experiment uses HPLC-Q-TOF-MS method to analyze the components of different parts of different varieties of Lily from the perspective of component analysis,and explore the component differences among five kinds of lilies;The bitter components in Longyahong lily were analyzed by column chromatography and nuclear magnetic resonance;Finally,colchicine and its 15 precursors were studied in five lilies,G.superba and C.autumnale.The following conclusions are drawn:(1)Firstly,the components of different parts of different varieties of lily were preliminarily identified by HPLC-Q-TOF-MS,and a total of 28 compounds were analyzed and identified,including:2 phenolic acids,7 glycosides,10 ketones,5 esters and 4sugars.The distribution of five lilies is:15 of Lanzhou lily`s leaves,14 of Lilium`s flowers,5of Longya lily`s leaves,3 of Longyahong lily`s bulbs,13 of Guiyanghong lily`s leaves.Among them,16 compounds were reported in lily for the first time.(2)Secondly,two monomer compounds were isolated from Longyahong lily by column chromatography and determined as the bitter component by sensory evaluation:the bitterness score of compound 3 was 3.4,slightly bitter;The bitterness score of compound 1is 4.2,more obvious bitter.The structure was identified by NMR,and the structural formula was obtained.Compound 3 was named:(25S)-spirostane-5-en-3β-27-dimethyl-3-O-[(1→2)-α-L-rhamnopyranosyl-(1→6)-β-D-glucopyranosyl]-β-D-glucopyranoside;Compound 1 is named:(22R,25R)-helix alcohol-5-en-3β-yl-O-β-D-glucopyranoside.(3)Finally,we used HPLC-Q-TOF-MS to detect colchicine in five different lilies,G.superba and C.autumnale.The experimental results showed that there was no colchicine([ESI~+],m/z=400.1755)and its precursor compounds in lilies,but colchicine and its 15precursor compounds could be detected in G.superba and C.autumnale.The above experimental results showed that there was no colchicine in lilies. |
