Chitosan oligosaccharides(COS)is a chitosan oligosaccharide with good biological activity,novel chemical structure and less adverse reactions.Compared with chitosan,chito-oligosaccharides have higher water solubility and lower viscosity,easier to be absorbed by passive objects,and higher bioavailability.Therefore,chito-oligosaccharides have a wider application prospect in health care,daily chemical and other fields.However,at present,the effects of chito-oligosaccharides on lipid metabolism are limited to oral administration,and the regulatory effects of extra-dermal chito-oligosaccharides on lipid metabolism disorders and the possible mechanisms have not been fully studied.In addition,as a natural raw material with various cosmetic effects,the application of chito-oligosaccharides in the field of daily use remains to be explored.Based on its ability to remove oil and oil and its strong inhibitory effect on a variety of bacteria,this paper has conducted a certain exploration on the evaluation of its efficacy in oil control and acne removal.Firstly,the basic characterization of chito-oligosaccharides was carried out,including the determination of reducing sugar content,miscellaneous protein content,ash content and molecular weight.Then take into account when cosmetics are used outside the skin,larger concentrations may have certain toxic side effects on organisms.Therefore,the toxic effect of chitosan oligosaccharide on HaCaT cells was explored.By observing the effect of different concentrations of chitosan oligosaccharide solution on the proliferation of HaCaT cells,it was found that the toxicity of chitosan oligosaccharide on HaCaT cells was minimal when the concentration range of chitosan oligosaccharide was 4/L.Then,the transdermal absorption of chitosan oligosaccharide and its thickening compound products were investigated to some extent.It was found that chitosan had negligible effect on the transdermal absorption of chitosan oligosaccharide products,and the optimal thickening concentration was 1%.After verifying the safety and effectiveness of chito-oligosaccharides for external use,the second part of the experiment used yeast cells as the carrier to explore its influence on microbial lipid metabolism.In order to achieve the best therapeutic effect,chito-oligosaccharide concentration of 4g/L was selected to explore the effects of chito-oligosaccharide on lipid production and lipid lowering pathways of yeast cells.Finally,it was concluded that chito-oligosaccharide had no obvious effect on lipid lowering of yeast cells,but had a good inhibitory effect on lipid production of yeast cells.After verifying that chito-oligosaccharides have a certain lipid metabolism effect on microbial cells,the third part of this paper uses mice as the carrier to conduct corresponding animal experiments to explore the regulatory effects of chito-oligosaccharides with different polymerization degrees on lipid metabolism in the body.The results showed that COS2-3 had little effect on lipid-lowering metabolism of normal mice,but had significant effect on lipid-lowering metabolism of obese mice.However,COS5-6 has a significant effect on the lipid-lowering metabolism of both normal mice and obese mice.The effects of chito-oligosaccharides on the contents of HDL-C,TC and TG in mouse serum are not regular,so it is speculated that the external application of chito-oligosaccharides does not affect the contents of HDL-C,TC and TG in mouse serum.Moreover,chitosan oligosaccharides can reduce fat by reducing the diameter of white adipose cells,so as to have a beneficial regulatory effect on lipid metabolism.Finally,the oil control and acne removal effects of chito-oligosaccharides and their derivatives were evaluated.The results showed that the MIC values of chito-oligosaccharide and zinc salt against propionibacterium acens were 0.5g/L and0.4g/L,and the MBC values were 0.8g/L and 0.6g/L,respectively.The inhibition rates of chitosan oligosaccharide and chitosan oligosaccharide zinc salt on 5α-reductase reached 67% and 83%.Chitosan oligosaccharides and their derivatives showed good oil control and acne removal effect,and can be used as cosmetic materials to improve the skin water and oil balance,to achieve the effect of acne treatment.
