| ObjectiveThis topic research plans through clinical trials,as IL-17A and IL-17E cytokines of IL-17 family for observation index,by using ELISA method and RT-PCR method to explore the correlations of IL-17A and IL-17E with URSA,discussing the expression of IL-17A and IL-17E after URSA patients taking Bushen Huoxue Prescription treatment,to observe the clinical curative effect of Bushen Huoxue Prescription treatmenting URSA,and then provide theoretical basis for clinical treatment of URSA.Materials and methodsPart one:The expression of IL-17A and IL-17E in the peripheral blood and IL-17R in decidual tissue with the URSA pregnancy failure patients.Collecting the peripheral blood and decidual tissue in 15 cases of the URSA pregnancy failure patients and 15 cases of normal pregnancy induced abortion which meeting the inclusion criteria,and extracting serum and decidual tissue under test.Applicating the ELISA method to detect the expression level difference of IL-17A and IL-17E in the peripheral blood of both patients.Applicating the RT-PCR method to detect the expression level difference of IL-17R mRNA in the decidual tissue of both patients.Part two:The expression of IL-17A and IL-17E in the peripheral blood with the URSA pregnancy patients by taking Bushen Huoxue Prescription for tocolysis treatment.Collecting 30 cases of the URSA tocolysis patients and 30 cases of normal pregnancy which meeting the inclusion criteria,getting the peripheral blood respectively both in URSA tocolysis patients before and after oral Chinese medicine treatment and in normal pregnancies when into the group before and after.Applicating the ELISA method to detect the expression level difference of IL-17A and IL-17E in the peripheral blood which both in the URSA tocolysis patients before and after oral Chinese medicine treatment and in normal pregnancies pregnancy into the group before and after.ResultThe first part shows that compared with normal pregnancy,the high IL-17A expression level and the low IL-17E expression level are present in the peripheral blood of URSA pregnancy failure patients,the IL-17R mRNA expression level is low in decidual tissue;The second part shows that compared with normal pregnancy control into the group before,in the serum of URSA group before treatment the expression level of IL-17A is more higher significantly,the expression level of IL-17E is more lower significantly;in the serum of Normal pregnancy control into the group after the expression level of IL-17A and IL-17E were significantly increased.URSA after treatment group,compared with before treatment,in the serum the expression level of IL-17A is significantly reduce and IL-17E is significantly rise;compared with normal pregnancy after groud,in the serum the expression level of IL-17A is significantly rise and IL-17E is significantly reduce.Conclusion(1)URSA decidua tissue of patients with low expression of IL-17mRNA associated with abortion.(2)URSA expression in peripheral blood of patients with IL-17A is significantly increased,the amount of IL-17A in favor of maintaining the pregnancy continued,resulting in overexpression of the Th17/Treg imbalance,is not conducive to the maintenance of pregnancy,it may be the cause of miscarriage one.(3)URSA in patients with significantly decreased the expression of IL-17E,the amount of IL-17E to help maintain the pregnancy continues,the low expression may result in Th1/Th2 balance disorders,maternal-fetal interface immune imbalance and abortion.(4)Bushen Huoxue Prescription-treated kidney deficiency and blood stasis URSA patients can be effectively reduced levels of IL-17A and IL-17E expression regulator increases,improve maternal-fetal immune tolerance,in order to continue the pregnancy to provide some protection.(5)The 30 cases of kidney and blood stasis URSA with Bushen Huoxue Prescription reatment is rate 83.33%clinical cure,improvement rate of 13.33%. |
