| Objection:To observe the effects of xiongfu san on the rat model of migraine induced with glycerol trinitrate(GTN).The rat ethology,the calcitonin-gene-related peptide(CGRP)and nitric oxide(NO)of contained in the aorta abdominalis blood are investigated to explore the mechanism of treating migraine and then provide factual basis on clinical popularization and application.Methods:The rat model of migraine-induced with glycerol trinitrate(GTN)is needed.72 healthy male SD rats were divided randomly and evenly into 4 groups:control group,model group,western medicine group and Chinese medicine group.Each group is divided randomly and evenly into 3 small groups by 3 different time points,1h,2h and 4h,so there are 12 small groups and 6 rats for each one.Every rat received gastric perfusion of medicine or distilled water once a day for 8 days continuously.Blank control group and model group is given 0.9%normal saline 1ml.100g-1,western medicine group is given 0.09mg.ml-1 Flunarizine solution 1ml.100g-1,Chinese medicine group is given 0.36mg.ml-1 xiongfu san 1ml.100g-1.On the 8th day,duplicate the expexperimental rat model of migraine induced with glycerol trinitrate(GTN)by neck hypodermic injection and observe The rat ethology,the calcitonin-gene-related peptide(CGRP)and nitric oxide(NO)of contained in the aorta abdominalis blood respectively after 1h,2h and 4h.Results:(1)Whthin 1h,the modle group’s(40.6±6.44)ethology grade is higher than control group(3.2±0.74)、western medicine group(12.56±5.68)and Chinese medicine group(10.47±4.30)and the results have the Statistical significance(P<0.05).(2)Control group(118.00±22、89116.21±10.20、109.42±13.34)and Chinese medicine group’s(114.14±21.24、96.12±14.40、115.39±13.11)CGRP means at 1h,2h and 4h are evidently lower than the modle group(179.78±19.85、185.49±31.28、253.86±31.52)and the results have the Statistical significance(P<0.05);western medicine group(124.38±17.13、138.80±15.53、82.04±17.64)and Chinese medicine group’s(114.14±21.24、96.12±14.40、115.39±13.11)CGRP means at lh,2h and 4h have no Statistical significance(P>0.05);western medicine group’s(82.04±17.64)CGRP means at 4h lower than the modle group(253.86±31.52)and have the Statistical significance(P<0.05);modle group’s CGRP(253.86±31.52)means at 4h evidently higher than at 1h(179.78±19.85)and and have the Statistical significance(P<0.05);western medicine group’s CGRP(82.04±17.64)means at 4h evidently lower than at 2h(138.80±15.53)nd have the Statistical significance(P<0.05);(3)After modling,control group(6.65±0.51、7.28±0.64、7.19±0.77)、western medicine group(11.52±1.03、9.60±1.02、9.06±1.12)and Chinese medicine group’s(10.93±2.30、10.76±1.23、8.82±0.81)NO means at lh,2h and 4h are lower than the modle group(20.71±4.02、15.04±0.94、12.27±0.81),have the Statistical significance(P<0.05);western medicine group(11.52±1.03、9.60±1.02、9.06±1.12)and Chinese medicine group’s(10.93±2.30、10.76±1.23、8.82±0.81)NO means at 1h,2h and 4h have no Statistical significance(P>0.05).The control group(5.00±0.422、4.91±0.333、5.33±0.422),western medicine group(5.67±0.490、5.25±0.447、5.63±0.522)and Chinese medicine group’s(5.27±0.333、4.80±0.490、5.47±0.333)aquaeductus mesencephali of dopamine D2 receptor expression levels at 1h,2h and 4h compared with modle group(5.33±0.433、4.67±0.422、5.50±0.500)have no Statistical significance(P>0.05).Groups at 1h,2h,4h three time points the aquaeductus mesencephali of dopamine D2 receptor expression levels have no significant difference(P>0.05).Conclusion:Xiongfu san can relieve migraine induced with glycerol trinitrate(GTN)by reduce the release of vasoactive substances CGRP and NO and improve dysfunction of the brain and blood. |
