Objective To study the effect and mechanism of Qiangguyin on periprosthetic osteolysis in rat which induced by polyethylene particles.Methods 40 adult male SD rats were randomly divided into blank control group,model group,Alendronate sodium treatment group and Qiangguyin Decoction treatment group,10 rats for each group.Blank group did not undergo any process,in model group,Alendronate sodium treatment group and Qiangguyin Decoction treatment group,polyethylene particles and titanium rods were implanted into the right femur,3 days after the completion of the model operation,in Alendronate sodium treatment group,rats were treated by Alendronate sodium,2ml Alendronate sodium(suspension of distilled water and tablets)once daily,with a dose of 0.083mg/(kg·d).in Qiangguyin Decoction treatment group,rats were administered 2ml volume drugs per 100 g through intragastric administration,three times a week.in blank group and model group,rats were administered 2ml volume saline per 100 g through intragastric administration,once daily.All the rats were fed in the same condition,and killed 12 weeks later,all experimental animals were sacrificed to prepare the bilateral femur specimens,Take the rat blood of inferior vena cava,blood was obtained to test serum TRAP5 b,TRAP staining and Lytic cytokines IL-1β,IL-6,TNF-α.take the rat right femur bone tissue to test the bone mineral density.Results There were no difference in the daily actions of rats between the 4 group during the course of experiment.The rat were in good mental states and rapid response and their body masses gradually increased.The incisions of rats in model group and Alendronate sodium treatment group and Qiangguyin Decoction group were healed at 1 week after the surgery.No rats died in each group during the experiment.Qiangguyin Decoction and Alendronate sodium can Reduce the level of serum TRACP5 b obviously,compared with model group,the two groups were statistically significant(P<0.05).through the observation and analysis of TRAP staining,the amount of OC in Alendronate sodium treatment group and Qiangguyin Decoction treatment group were less than that in model group(P<0.05).the Concentration of IL-1β、IL-6、TNF-α in the two group was lower than that in model group(P<0.05).the bone mineral density in the two group was increased than that in model group(P<0.05).Conclusions Qiangguyin Decoction can effectively inhibit the process of the periprosthetic osteolysis and increase bone mineral density in model rats,its mechanism may be to reduce inflammatory reaction and inhibit the activity of OC,inhibit bone resorption around the prosthesis.
