Prunus Flos are the dried flower buds of Prunus mume(Sieb.)sieb.et Zucc.members of the rose family.The chemical components of Prunus Flos were rich,among which the flavonoids represented by hypericin and phenylpropanoids represented by chlorogenic acid were higher.The ethanol extract of Prunus Flos has obvious anti-depression effect on mice,and the total flavonoids of Prunus Flos can significantly improve the depression behavior induced by CUMS in rats.On the basis of this study,in order to find monomer compounds with biological activity.Objective: To enrich the chemical constituents of Prunus Flos by screening the extraction activities of different parts of Prunus Flos and systematically separating the active parts,so as to search for monomer compounds with biological activities.Methods: Corticosterone-induced SH-SY5 Y cell model was used to screen the activity of different extracts from Prunus Flos.The active parts were separated and purificated systematically by using a variety of chromatographic separation technology(silica gel,MCI,Sephadex LH-20,Rp-8 column chromatography,high efficiency liquid preparation,etc.)and using the modern spectral techniques to identify the structure of the isolated monomer compounds.Results: 20 compounds were isolated and purified from Prunus Flos,and 15 of them were identified as benzoic acid(1),pinoresinol(2),naringin(3),isorhamnetin(4),trans-ferulic acid(5),quercetin(6),kaempferol(7),ethyl-β-D-glucopyranoside(8),mumeic acid-A methyl ester(9),mumeic acid-A(10),quercetin-3-O-β-D-glucopyranoside(11),astragalin(12),Isoquercitrin-6′′-O-benzoate(13),hypericin(14),rutin(15).Compounds 2,3,4,6,7,12,13,14 and 15 were screened for their protective effects on corticosterone-induced SH-SY5 Y cells.The results of MTT assay showed that all the above compounds could significantly increase the cell survival rate.Conclusion: The extracts of ethyl acetate and n-butanol from Prunus Flos had certain protective effects on corticosterone-induced SH-SY5 Y cells.Among the compounds isolated from the ethyl acetate site,compounds 13 was a new natural product,compounds 2,3,8 and 10 were firstly isolated from the Prunus Flos.Compound2,3,4,6,7,12,13,14 and 15 had protective effects on corticosterone-induced SH-SY5 Y cells. |
