Rosae Chinensis Flos has a long history of cultivation and is distributed in Hebei,Shandong,Hunan,Yunnan,Sichuan and Xizang.The 2015 edition of Chinese Pharmacopoeia records that Rosae Chinensis Flos has sweet taste and warm sex.Liver meridian.It has the effect of activating blood circulation and relieving depression.For qi stagnation and blood stasis,irregular menstruation and thoracolumbar distension pain.Literature studies have shown that flavonoids are the main active chemical components of Rosa chinensis Jacq.with antibacterial,antiviral,antioxidant,anti-tumor and hypoglycemic pharmacological effects.Rosa chinensis Jacq.is cultivated everywhere and is rich in resources.Literature research found that there has been no relevant report on the coagulation activity of Rosae Chinensis Flos.so this topic studied the chemical composition and coagulation effect of Rosae Chinensis Flos.The first chapter summarized the research progress of chemical constituents and biological activities of Rosae Chinensis Flos;Chapter 2 studied on coagulation activity and chemical constituents of Rosae Chinensis Flos;Chapter 3 studied on the mechanism of procoagulation activity of astragalin in Rosae Chinensis Flos.Firstly,the research progress of chemical components and their biological activities of Rosae Chinensis Flos since 2000 was reviewed,and provided a theoretical basis for the development of the chemical constituents and coagulant activities of Rosae Chinensis Flos.Secondly,the coagulant activity of different extracted parts of Rosae Chinensis Flos was evaluated by the screening of coagulation factors in vitro.The results showed that the total extract of Rosae Chinensis Flos could significantly prolong APTT,significantly prolong PT and reduce FIB,indicating that the total extract of Rosae Chinensis Flos had very good anticoagulant activity,and the effect was equivalent to that of breviscapine.The ethanol fractions of 20%,40%,60% and 95% could shorten PT remarkably,indicating that the ethanol fractions of 20%,40%,60% and 95% had good effect on promoting blood coagulation,the effect of other fractions was similar to that of Yunnanbaiyao.The total extract of Rosae Chinensis Flos was isolated by various chromatographic techniques by bioassay-guided Method.Their structures were identified by NMR.15 compounds were identified including kaempferol(1),quercetin(2),isoquercitrin(3),astragalin(4),afzelin(5),quercitrin(6),phenylethyl glucopyranoside(7),avicularin(8),juglanin(9),nicotiflorin(10),phenylethyl-6'-O-galloy-?-D-glucopyranside(11),tiliroside(12),methyl gallate(13),8-O-methylherbacetin-3-O-?-D-sophoroside(14),gallic acid(15).Compounds 8,10,13 and 14 were isolated from the plant for the first time.Thirdly,compounds were screened for coagulation activity in vitro,and the results showed that compound 4(astragalin)could significantly shorten APTT and TT,and significantly increase FIB,indicating that astragalin had very good procoagulant activity,and the effect was similar to that of Yunnanbaiyao.Compound 12(tiliroside)could shorten PT significantly and had good procoagulant activity.Finally,the mechanism of procoagulant activity was investigated for high content of astragalin in vivo.The time of coagulation was significantly shortened in both groups compared with the model group.The results of the canthus blood collection microscopy showed that astragalin(10 mg/kg)significantly increased the number of platelets and had a very significant procoagulation effect(P < 0.001).In terms of the screening of coagulation factors in vivo.Compared with the model group,both the positive group and the model group(10 mg/kg)could significantly reduce PT and APTT(P < 0.001),and significantly increase the concentration of FIB(P < 0.01)to play a role in promoting coagulation.By measuring the content and ratio of TXB2 and 6-Keto-PGF1 a in rats,it was speculated that the effect of TXA2/PGI2 on promoting blood coagulation was regulated.Through the determination of e NOS and ET-1 contents in rats,it was speculated that astragalin might promote blood coagulation by regulating the levels of NO and ET in rats.In addition,astragalin could also increase the whole blood viscosity and plasma viscosity of rats,and increase ESR and PCV in rat plasma to play a role in promoting blood coagulation.
