Objective To invest gate the relationship between monocyte chemoattractant protein-1(MCP-1),high mobility group box 1(HMGB1)and matrix metalloproteinase9(MMP-9)with preterm premature rupture of membranes(PPROM).Methods There were 216 pregnant women who were birthing babies in the obstetrics department of Yanbian University since January 2017 to December 2019.102 pregnant women with preterm premature rupture of membranes were divided into with chorioamnionitis group that includes 34 women and without chorioamnionitis group that includes 68 women.At the same time,114 full-term pregnant women who delivered in the Yanbian University Affiliated Hospital were selected as the control group.The concentration of MMP-9 in umbilical cord blood was detected by enzymelinked immunosorbent assay while the concentration of MCP-1 in umbilical cord blood was detected by flow cytometry.The level of MMP-9 in placenta was determined by Western blotting and the level of HMGB1 in placenta was detected by immunohistochemical staining.The whole genome was extracted from whole umbilical cord blood.MMP-9 rs3918242 was amplified by polymerase chain reaction.The genotype of the target site was determined by Sanger sequencing.Results 1.The concentration of MMP-9 in cord blood of the control group was 503.21 ± 123.64 pg/mL,while that of the PPROM group was 597.54 ± 171.94 pg/mL.The content of McP-1 in cord blood of the control group was 129.65±50.91pg/mL,while the content of MMP-9 in cord blood of the PPROM group was 146.23±63.82pg/mL.The levels of mmp-9 and McP-1 in umbilical cord blood were not significantly different between the two groups(P>0.05).2.In the PPROM group,the content of MMP-9 in umbilical cord blood of parturient women with chorioamnionitis was 775.73±101.64 pg/mL,and that of the group without chorioamnionitis was 543.62±93.42 pg/mL.McP-1 content in umbilical cord blood of pregnant women with chorioamnionitis was 170.53±31.67 pg/mL.3.The positive rate of placenta HMGB1 in PPROM group was higher than that in control group(P<0.05).Compared with the control group,PPROM maternal placenta MMP-9 levels were higher.4.In PPROM group,CC genotype accounted for 47.1%,CT genotype 49%,TT genotype 3.9%;In the control group,CC genotype accounted for 73.7%,CT genotype 21%,TT genotype 5.3%.In PPROM group,the proportion of CT genotype was higher than that of CC genotype,while in control group,the proportion of CC genotype was higher than that of CT genotype.The genotype frequency and allele frequency of MMP-9 RS3918242 in PPROM group and control group were different(P<0.05).Conclusion 1.The high levels of placenta MMP-9 and HMGB1 are related to PPROM.2.The concentration of MMP-9 and MCP-1 in umbilical cord blood related to PPROM combined with chorioamnionitis.3.MMP-9 rs3918242 single nucleotide gene polymorphism is related to PPROM.The C allele may be a protective factor of PPROM,and the T allele may be a risk factor of PPROM.
