Fruits of physalis alkengi L.is an herbaceous plant of Solanaceae,which has the functions of clearing heat,relieving sore throat,relieving internal heat or fever,eliminating phlegm and diuresis.It’s fruit usually used as medicine together with it or persistent calyx.In addition to the functions of clearing heat and relieving internal heat or fever,the fruit of physalis alkengi L.has also attracted the attention of educational circles in its functions of reducing blood fat and sugar,which is a promising traditional Chinese medicine for both food and medicine.Because of the changes in the social life style,the incidence rate of hyperlipidemia and other metabolic diseases has also increased.Therefore,this study is aimed at separating and identifying the chemical constituents of the fruit of physalis alkengi L.,and trying to use the lipid-lowering experiment to study the effective parts of the fruit,so as to explore its effective substance foundation and lay a foundation for further development of new drugs and other research.The fruits of physalis alkengi L.was extracted twice with 75%ethanol under reflux.The extracts were combined and recovered under reduced pressure to obtain an extract,which was then eluted through the macroporous AB-8 and macroporous resin to obtain 4 parts,namely WE,30%EE 70%EE,95%EE.70%EE compounds were separated by repeated silica gel column chromatography,thin layer chromatography,PHPLC and other separation methods until monomer compounds were obtained.Then using mice to conduct high-fat experiment to evaluate the effective part of lipid-lowering effect of the fruit of physalis alkengi L..8 groups of mice were adaptively dosed 7 days(with Xuezhikang as the positive control,the rest of them were tested for each group,the blank group and the model group were given normal saline),except the blank control group,all the other groups were intraperitoneally injected with Yolk Emulsion.After 24 hours,the blood samples were taken from the abdominal cavity.Centrifugation was used to determine the values of TC and TG in the serum,and compared with the model group.Finally,seven monomers were isolated,including Ginsenine(1),26-O-β-D-glucopyranosyl-3β,20α,26-triol-25(R)-Δ5,22-diene-furost-3-O-α-L-pyran rhamnosyl(1→2)-[α-L-pyranorhamnosyl(1→4)]-β-D-glucopyranoside(2),3-O-α-L-arabinopyranose-Ivy saponin-28-O-(4-O-acetyl)-α-L-rhamnopyranose-(1→4)-β-D-glucopyranose-(1→6)-β-D-glucopyranoside(3),2α,3β-dihydroxy-5α-pregn-16-en-20-one-3-O-β-D-glucopyranosyl-(1→4)-β-D-galactopyranoside(4)and 23-hydroxy-gitogenin-3-O-β-D-glucopyranosyl-(1→4)-β-D-galactopyranoside(5),Sucrose(6),maltose(7).Through the high fat experiment of mice,we could find that:Compared with the model group,the blank group,positive drug group and 70%EE,WE group could significantly reduce the content of TC and TG(p<0.05).There was no significant difference in the Total extract group,30%EE group and 95%EE group.By means of literature search,compounds 1-5 were isolated from the the fruit of physalis alkengi L.for the first time,among which compound 5 was a new compound retrieved by Scifinder database.The extracting part of the fruit of physalis alkengi L.with 70%ethanol significantly reduce the serum TC and TG of mice,and the water extract part also had a certain effect(p<0.05).
