Objective: The rabbit model of steroid-induced ischemic necrosis of the femoral head was established by different doses of lipopolysaccharide combined with glucocorticoid methylprednisolone.Methods:Using a randomized controlled animal experiment,30 rabbits in the Guangxi Medical University Animal Center feed(maintaining humidity,temperature,ray,interference)adaptive feeding.During the feeding period,food and water can be placed in the position of the rabbit straight position,which can cause each group of experimental animals to eaten or drink water.Feeding a week after a week(6 each): Group A: 5μg / kg lipopolysaccharide group;Group B: 10μg / kg lipopolysaccharide group;Group C Group: 20 μg / kg lipopolysaccharide group;D Group: Simple hormone group;Group E: Blank control group.For each rabbit,the left hip muscle injection of 100,000 U / kg penicillin prevents antiinfection.After 24 h,the two groups group C,group D,and the other side hip muscles,the right hip muscle injection.20 mg / kg of glucocorticoid methylprednisolone,blank control group E group of normal saline in the right side hip muscle injection.After 48 hours,the rabbits were intravenously intravenous by the rabbits: A group of ear veins of LPS(5 μg / kg),B group vein injection LPS(10 μg / kg),c group ear vein injection LPS 20 μg / kg),D group ear intravenous injection,etc.,physiological saline,E group of ear veins,etc.,is also injected with volume saline.After the experimental rabbit LPS was injected,the experimental rabbit activity of each group of each group was observed after 6hours after injection of LPS.Injecting LPS after 24 hours,the left,B group,C group,and the D group were left in the gluteus,and the right hip muscle alternately injected with methylprednisolone(20mg / kg),with a total injection of24 hours interval.2 times.After the end of the last glucocorticoid methylprednisolone,the muscle injection of penicillin(100,000 u / kg)was used to prevent anti-infection,and the left and right hip muscles were alternately injected with 48 hours each time,the duration was 4 weeks.After 4 weeks,the survival,health ratio,prevalence,mortality,MRI examination results of each group were observed,and the results of histopathological slices were observed,and the necrosis rate and spontaneous necrosis of rabbit femoral heads and sputum depression were calculated.Results: 1.The survival rabbits in each group is a group of 5 in group A,and the B group is survive.The C group is survived.The D group is survived,and the E group 6 survives.2.With the increase in injection LPS injection,the activity of rabbits is significantly inhibited;the state of the active state of the simple hormone group and the blank control group is not affected.3.There is no significant difference in the health of rabbits,prevalence and mortality statistics.4,each group of rabbit femoral head ischemic necrosis MRI examination results Tips:Group B,Group C,D groups have irregular low signals under T1W1,E group has no abnormal low signal under T1W1;group A,Group B,C groups can be seen in T2W1,irregular high signals,and there are abnormal low signals around,and the D group and E have no significant abnormal signals.5,each group of rabbit femoral head tissue pathological sections(HE dyeing): group B,group C,group C,E group bone smaller,increased empty bone trap,fat cell hypertrophy;Group B,C group increases the bone lamination of the LPS injection dose,and the number of empty bone trap has increased,and the fat cells have increased fat.6,C-group femoral head ischemic necrosis,the necrosis rate is the highest,the D group is the lowest death rate,A,B group die is in the middle,and the E group will not see necrosis.The fracture rate of bone bones in each group is as follows:Group A,and the left and right femoral head of the bone in group B is significantly higher than that of the D group;group A and the right femoral bone in the right femoral head in group group A is higher than The left femoral head is branched by the bone,but there is no significant difference;the right femoral head cascade in the C group was significantly higher than the left femoral head.Conclusion: When the molding of the mold LPS injection is 20 μg / kg,10 μg / kg and 5 μg / kg,respectively,with the increase of the lipopolysaccharide injection dose,this modeling method has this modeling method without affecting the survival and mortality of the experimental animal rabbit Sustainable safety and reliability;However,the injection dose of the fat polysaccharide is related to the degree of ischemic necrosis of femoral head,the higher the dose,t he higher the femoral head of the femoral head,the more obvious the degree of femoral necrosis.
