| Objective:Observe the effect of Yimu Suogong granules on the level of endothelin and inflammatory factors in uterine tissue homogenate of early pregnancy incomplete abortion rats.Eexplore the possible mechanism of abnormal vaginal bleeding after medical abortion,to provide theoretical basis for treatment of the disease.Methods:Select sexually mature SD female and male rats(2 months,48 female rats and 20 male rats),Eight female SD rats were randomly selected as a blank group after adaptive feeding in separate cage.female rats: male rats =2:1were caged at 18:00 PM,observe whether there is Yin suppository or vaginal secretions microscopic examination revealed sperm at 08:00 next morning.Rats were considered as the first day of pregnancy if so,and the non-pregnant rats continued to be caged until Yin suppository or sperm were found.Mifepristone and misoprostol were gavaged on day 7 of gestation,after administration of misoprostol,a sterile cotton ball of suitable size was inserted into the vagina,and if the cotton ball was found blood,the mold was considered successful in the next morning.Successful models of rats were divided into model group,Yimu Suogong granules low-concentration group,medium-concentration group,high-concentration group and Xinshenghua granules group,with 8 rats in each group.Blank group and model group were given drinking water of 1ml/(100g/d)for consecutive 7 days.Yimu Suogong granules low-concentration group,medium-concentration group,and high-concentration group were given Yimu Suogong granules at low dose(1.56g/kg.d),medium dose(3.13g/kg.d)and high dose(6.25g/kg.d)for consecutive 7 days.Xinshenghua granules group was given xinshenghua granules(1.56g/kg.d)for 7 consecutive days.The rats were anesthetized and sacrificed on the 8th day after the group was enrolled.The uterus tissues of rats were quickly found and dissected after laparotomy,then observed the uterine tissue morphology and pathological changes.Use elisa determining the level of ET-1,IL-6 and TNF-α in uterine tissue homogenate and determining the level of NO by nitrate reduction method.Results:(1)Uterine pathological grades of rats:Uterine pathological grades of blank group was significantly higher than that of model group,the difference was statistically significant(P<0.01),what confirmed the model of incomplete abortion was successfully established.The uterine pathological grades of Yimu Suogong granules low-concentration group,medium-concentration group,and high-concentration group were significantly higher than model group,with statistical significance(P < 0.05).Among them,Yimu Suogong granules low-concentration group,medium-concentration group,and high-concentration group had the same efficacy as Xinshenghua granules group in improving the uterine pathological grades,with no statistical significance(P >0.05).(2)Compared with model group,Yimu Suogong granules could increase the content of ET-1 in uterine tissue homogenate of rats,with statistical significance(P<0.01).Compared with Xinshenghua granules group,there was no significant difference in the content of ET-1 in uterine tissue homogenate between Yimu Suogong granules medium-concentration group and high-concentration group(P=0.953,P=0.869).Compared with model group,NO content in uterine tissue homogenate of rats in Yimu Suogong granules medium-concentration group,high-concentration group and Xinshenghua granules group were significantly decreased,with statistical significance(P<0.01),while there was no statistical difference in Yimu Suogong granules low-concentration group(P>0.05).NO content in uterine tissue homogenate of Yimu Suogong granules medium-concentration group,high-concentration group and Xinshenghua granules group showed no statistical difference(P=0.295,P=0.937).(3)Compared with model group,Yimu Suogong granules could reduce the IL-6 content in uterine tissue homogenate of rats,the difference was statistically significant(P<0.01),Compared with Xinshenghua granules group,the IL-6 content in uterine tissue homogenate of Yimu Suogong granules low-concentration group,medium-concentration group showed no statistical difference(P=0.250,P=0.125),the IL-6 content in uterine tissue homogenate of Yimu Suogong granules high-concentration group showed statistical difference with Xinshenghua granules group(P=0.013).(4)Compared with model group,Yimu Suogong granules could reduce the TNF-α content in uterine tissue homogenate of rats,the difference was statistically significant(P<0.05).Compared with Xinshenghua granules group,there was no significant difference in TNF-α content in uterine tissue homogenate of Yimu Suogong granules low-concentration group and medium-concentration group(P=0.141,P=0.128),but there was difference in TNF-α content in uterine tissue homogenate of Yimu Suogong granules high-concentration group(P=0.036).Conclusion:(1)Yimu Suogong granules can improve the pathological grades of uterus in rats with incomplete medical abortion,reduce the inflammatory response of endometrial tissue,promote the excretion of residual villi and decidua tissue and promote endometrial involution.(2)Yimu Suogong granules can increase the content of ET-1 and reduce the content of NO in rat uterine tissue homogenate,confirmed that the mechanism of Yimu Suogong granules reduced abnormal uterine bleeding after medical abortion may be related to adjust the balance of ET-1 and NO and accelerat the contractile repair of uterine muscle tissue and blood vessels.(3)Yimu Suogong granules can reduce the levels of NO 、 IL-6and TNF-α in uterine tissue homogenate of incomplete abortion rats,decrease the expression of inflammatory factors,which may be related to treatment of abnormal uterine bleeding after drug flow.(4)The efficacy of Yimu Suogong granules and xinshenghua granules in improving the pathological grades of uterus is similar,but it is more effective in improving inflammatory factors and reducing inflammation which can reduce the risk of infection after medical abortion and be popularized in clinical treatment of related diseases. |
