Objective:Through animal experiments to explore the effects of Yimu Suogong Granules on the PGI2 / TXA2 system and coagulation function of rats with incomplete abortion,in order to explore the mechanism of Yimu suogong granules in preventing vaginal bleeding after drug abortion.Methods:Sexually mature SD rats were randomly selected to take 8 female rats as blank control group(group A)after a week of adaptive feeding,and then the rest rats were caged according to the male-to-female ratio of 1: 2,check the female suppositories or vaginal secretion smears in the next morning to determine whether the rats were pregnant.The pregnancy was confirmed and included in the experiment.The day of pregnancy was recorded as d1.If it was not conceived,it continued to be caged at 18:00 for one week.Eight rats were randomly selected as the pregnancy control group(Group B).The other pregnant rats(47 rats)were given mifepristone(8.3 mg/kg)+ misoprostol(100?g/kg)for gavage on day d7 of pregnancy,both groups A and B were treated with drinking water instead of drugs,and one cotton ball was placed in the vagina after the administration of misoprostol.Obvious blood stains were considered as successful modeling,and 44 successful rats were randomly divided into the model group(group C)8 and theYimu Suogong Granules group(group D 27,of which the low-dose group D1 and medium-dose GroupD2 group,high-dose group D3 group 9 each),XinShenghua granule group(group E)9 rats.Rats in each group started intragastric intervention drugs at d8 of conception.D1,D2,and D3 groups were given 0.313 g/mL,0.625 g/mL,and1.25 g/mL of Yimu Suogong Granules suspension at 10 ml/(kg.d).Group E was given 0.281 g/mL of the suspension of XinShenghua granules at 10 ml/(kg.d).The rest of the groups were replaced with drinking water for 7 days.Anesthetized and anaesthetized the rats at d15 during pregnancy.Blood was taken from the abdominal aorta,part of the blood was taken to separate the serum,and the serum 6-keto-PGF1α and TXB2 concentrations were measured by Elisa method.The other part of blood is separated after plasma anticoagulation treatment to detect PT,TT,FIB and APTT values.The uterine specimens were taken for HE staining,and histological changes were observed with pathological scores.Results:(1)(1)Vaginal bleeding time: The vaginal bleeding time of rats in group D was significantly shorter than that in groups C and E(P <0.01).(2)Uterine wet weight: Groups C,D,and E were significantly reduced compared to group B;and groups D3 and E were significantly reduced compared to group C(P <0.05).(3)In terms of histopathology:the endometrium of the D and E groups showed different degrees of repair compared with the C group,of which the D3 group had better repair;compared with the C group,the chorionic decidual residual scores of the D3 and E groups were reduced significantly(P<0.01).(2)PGI2/ TXA2 system: Compared with group A,there was no significant difference in group B about TXB2,but 6-keto-PGF1α increasedsignificantly,and 6-keto-PGF1α/TXB2 increased,the difference was statistically significant(P<0.05);And comparing group B and group C,TXB2 in group C increased significantly,6-keto-PGF1α decreased,and 6-keto-PGF1α/TXB2 decreased significantly(P <0.05);Compared with group C,TXB2 in groups D1,D2,and D3 decreased(P<0.05),there was increased in 6-keto-PGF1α/TXB2 in groups D2,D3,E,and the difference was statistically significant(P <0.05).(3)Coagulation function: compared with group A,there were no statistical differences in PT,APTT,FIB,TT in group B,and the PT in group C was significantly prolonged(P<0.05);compared with group C,the PT in groups D1,D2,D3 and E was prolonged(P <0.05),and there was no significant statistical difference in FIB in groups D1,D2,D3,and E.The TT value in groups D1 and E was prolonged,and the APTT in group E was significantly prolonged.The difference were all statistically significant(P <0.05).Conclusion:Yimu Suogong Granules can effectively shorten the vaginal bleeding time in rats with incomplete abortion.(2)The imbalance of TXB2/6-keto-PGF1α is related to vaginal bleeding after drug incomplete abortion.Yimu Suogong Granules can adjust the ratio of TXB2/6-keto-PGF1αto reduce the TXB2 content of the model rat blood.Yimu Suogong Granules plays a role in restore the balance of PGI2/TXA2 state in reducing the vaginal bleeding time of model rats.(3)Yimu Suogong Granules can prolong the PT value of model rats,strengthen anticoagulation and promote the dissolution of blood clots on model rats,so that “qi” and blood can run smoothly.It is biochemical and has the characteristics of traditional Chinese medicine of "hemostasis without stasis".(4)Yimu Suogong Granules can promote the discharge of residual villi and decidua of the model rat uterus,promote the endometrium repair of the model rat and reduce the uterine pathological score.It is better to reduce the content of TXB2 in the blood of model rats than XinShenghua granules.For patients with "blood stasis syndrome",Yimu Suogong Granules may be preferred.
