Effect Of Zhuang Experience Prescription Wei Du Qing On Expression Of PTGS2/IL6/ICAM1 In Gastric Cancer Model Nude Mice

Objective: This study aims to explore the mechanism of action of Zhuang prescription Weiduqing on gastric cancer by using network pharmacology combined with GEO database analysis,Polymerase chain reaction(PCR),Western Blot(WB)and other experimental techniques,and preliminarily evaluate the effect of Weiduqing on gastric cancer.It provides objective scientific basis for the clinical popularization and application of Zhuang experience prescription Wei Du Qing.Methods: 1.Supplement the ingredients of "Zhuang experience prescription Wei Du Qing" from TCMSP,TCMID and other drug databases and literatures,screen the active ingredients and predict the molecular targets according to the pharmacokinetic properties.Gene chips related to gastric cancer were retrieved from GEO database for differential gene analysis.The intersection points of the two are drawn into a Venn diagram to generate a network map of drug disease targets.Cross genes were obtained from online STRING database to obtain PPI core protein interaction network.Cytoscape3.7.2 software was used for topological analysis,and R software was used for GO and KEGG enrichment analysis.2.Sixty male nude mice were randomly divided into 5 groups:model group,cisplatin positive control group(cisplatin group),gastric toxicity high,medium and low dose groups,with 12 mice in each group.Gastric cancer model was established by subcutaneous injection of GASTRIC cancer SGC-7901 cell suspension in nude mice.Weiduqing was given orally for 21 days,and cisplatin was injected intraperitoneally for 3 times(once a week).The tumor was weighed and the tumor inhibition rate was calculated.The m RNA and protein expressions of PTGS2,IL6 and ICAM1 were detected by PCR and WB.Results: 1.Network pharmacology analysis showed that a total of194 predictable active ingredients were retrieved from Zhuang experience prescription Wei Du Qing,including 35 core targets of "Zhuang experience prescription Wei Du Qing-Gastric cancer-target",including IL-6,ICAM1,CCL2,PTGS2,PRKCB,MMP9,CXCL2,VCAM1,etc.The KEGG pathways related to the treatment of gastric cancer by Wei Du Qing recipe include TNF signaling pathway,AGE-RAGE signaling pathway in diabetic complications,Fluid shear stress and atherosclerosis,Lipid and atherosclerosis,Micro RNAs in cancer,IL-17 signaling pathway,Amoebiasis,etc.The main biological processes involved in Zhuang experience prescription Wei Du Qing against gastric cancer include response to lipopolysaccharide,response to molecule of bacterial origin,vascular process in circulatory system,response to oxygen levels,cellular response to drug,response to nutrient,response to oxidative stress and so on.2.In terms of tumor inhibition rate,compared with the model group,the tumor volume inhibition rate and mass inhibition rate in the western medicine group,the high-dose,medium-dose and low-dose zhuang medicine gastric toxin qingqing groups were reduced to varying degrees(P<0.05).Compared with the tumor growth in the model group,the tumor growth in the high-dose,medium-dose and low-dose Zhuang experience prescription Wei Du Qing groups and cisplatin groups was relatively slow.Tumor growth inhibition was relatively significant in We Du Qing medium-dose group and cisplatin group(P < 0.05).In terms of protein expression,Western Blot results showed that Zhuang experience prescription Wei Du Qing could reduce the protein expression levels of PTGS2,IL6 and ICAM1 in gastric cancer nude mice(P < 0.05).In terms of gene relative expression level,PCR results showed that Zhuang Yiweiduqing Prescription could also reduce the m RNA relative expression levels of PTGS2,IL6 and ICAM1 in gastric cancer nude mice(P < 0.05).Conclusion: 1.Joint through the network pharmacology GEO genetic variations of Zhuang experience prescription Wei Du Qing treatment for gastric cancer mechanism analysis shows: the party through multiple targets,multiple channel ACTS on the stomach,anti-inflammatory,inhibiting oxidative stress,mainly through regulating immune mechanisms such as gastric cancer treatment,for Zhuang experience prescription Wei Du Qing mechanism provides a theoretical reference for the treatment of gastric cancer.2.This study showed that Zhuang experience prescription Wei Du Qing had tumor suppressive effect on transplanted gastric cancer nude mice,significantly inhibited tumor growth,and decreased m RNA and protein expression levels of PTGS2,IL6 and ICAM1,which may be regulated and control to the inhibition of TNF signaling pathway.
