Objective: To study the potential active ingredients and mechanism s of Longli Pingchuan Decoction in the treatment of childhood asthma through network pharmacology and molecular docking methods.Methods: TCSMP was used to screen the effective ingredients and targets of Longli Pingchuan Decoction;the Gene Cards database was searched to screen the genes related to childhood asthma and the target genes were obtained by intersection with the target;and the active ingredientsand targets of Longli Pingchuan Decoction were constructed using cytoscape 3.6.1 software The relationship network between genes and traditional Chinese medicines;through network topology analysis,screen the core targets of ginkgo biloba for treating childhood asthma,construct a protein interaction(PPI)network,and perform KEGG and GO enrichment analysis on target genes;use molecular docking technology to verifydrug components Combination with target;predict miRNA upstream of core gene,and construct miRNA-mRNA regulatory network.Results: 194 active ingredients were screened out from Longli Pingchuan Decoction to affect 207 target genes of childhood asthma;10 core compounds were quercetin,luteolin,kaempferol,wogonin,7-methoxy-2-Methyl isoflavones,stigmasterol,licorice chalcone A,β-sitosterol,ba icalein,naringenin;20 core targets are AKT1,TP53,JUN,VEGFA,CASP3,IL1 B,STAT3,ESR1,PTGS2,MAPK3,EGFR,HIF1 A,MMP9,MYC,EGF,FOS,CCND1,PPARG,FN1,CXCL8;enrichment analysis shows that GO of target genes is mainly rich in Nuclear receptor activity,RNA polymerase II transcription factor binding,ubiquitin-like proteinlig ase binding,protein phosphatase binding,cytokine receptor binding,DNA binding transcription activator activity,MAP kinase,cytokine activity;involving IL-17,TNF,PI3K-Akt,Th1/Th2 cell differentiation,Th17 cell differentiation,MAPK,p53,Toll-like receptor,HIF-1 and othersignalin g pathways.The 19 core genes were screened to have 10 upstream miRNAs in bronchial tissues;miRNA-mRNA network analysis showed that hsa-miR-7-5p,has-miR-126-3p,and hsa-miR-212-3p aremore common in childhood asthma important.Conclusion:(1)Longli Pingchuan Decoction in the treatment of childhood asthma is coordinated by multiple components,multiple targets and multiple pathways;(2)The results show that quercetin,luteolin,kaem pferol,wogonin,7-methoxy-2-Methyl isoflavones and other Longli Pingchuan Decoction’s main effective compounds achieve anti-inflammatory an d inhibit airway remodeling effects by inhibiting the conduction of sign al pathways at all levels;(3)Research shows that AKT1,TP53,JUN,VEGFA,CASP3,etc.may be possible is the key target gene of Longli Pingchuan Decoction in the treatment of childhood asthma;(4)Longli Pingchuan Decoction plays a role in the treatment of childhood asthma by regulating IL-17,TNF,PI3K-Akt,Th1/Th2 cell differentiation,Th17 cell differentiation,and other signaling pathways.
