Objective:Based on the traditional Chinese Medicine Inheritance assistance platform(TCMISS,V2.5),the herbal prescription rules of Professor Liu Ming in the treatment of chronic venous insufficiency(CVI)was studied,the mechanism of the core prescription in the treatment of CVI was explored by network pharmacology,so as to facilitate the experience inheritance of distinguished veteran doctors of traditional Chinese medicine(TCM).It also provides theoretical basis for TCM treatment of CVI.Methods:This study is divided into two parts.The first part is data mining.The TCM prescriptions of 515 patients with chronic venous insufficiency treated in the outpatient department by professor Liu Ming were collected,and an Excel database was established,which was standardized and recorded into the TCM Inheritance assistance platform(V2.5).Software integrated data mining methods are used to analyze the database.The second part is network pharmacology study.TCMSP database was used to search the active ingredients and predict the target of Traditional Chinese medicine,genes related to CVI were searched in OMIM and Dis Ge NET database,and target names were standardized using Uni Prot database.Draw a Venn diagram of the intersection targets.A drug-active component-target-disease network was constructed by Cytoscape 3.7.1 software.PPI network diagram was prepared by String database.GO functional enrichment analysis and KEGG pathway enrichment analysis were performed using DAVID database.Results:1.A total of 515 outpatient prescriptions of patients with CVI treated by Professor Liu Ming were collected in this study,among which 88 prescriptions contained symptom information.There were 38 cases of qi stagnation and blood stasis syndrome,24 cases of damp-heat syndrome and 26 cases of qi and blood deficiency syndrome.The 5most frequent symptoms were lower limb swelling,lower limb blue vein exposure,white moss,reddish tongue,thin white moss.A total of 192 Chinese herbs were used,and the 5drugs with the highest frequency were Licorice root,Red peony root,Tuckahoe,Angelica sinensis and Rhizoma alismatis.Drugs with four qi as "cold",drugs with five tastes as "gan" and drugs classified as "spleen" appeared in the highest frequency.2.Professor Liu Ming used 13 core drugs for the treatment of chronic venous insufficiency of lower limbs,which were "Poria cocos,Grifola,Rhizoma alismatis,Rhizoma ligustici walli-chii,Angelica,Red peony root,Licorice,Cohosh,Kudzu root,Astragalus membranaceus,Dangshen,Achyranthes bidentata and Cassia twig".3.Network pharmacology.A total of217 active ingredients and 154 action targets were collected from the core prescription,and1275 targets of CVI were collected.There were 47 intersection targets of active ingredients and diseases.After enrichment analysis,221 items related to biological processes,32 items related to cell composition,36 items related to molecular function,and 68 signaling pathways were obtained.Conclusion:1.Based on the analysis of professor Liu Ming ’s prescription for diagnosis and treatment of chronic venous insufficiency,it is concluded that professor Liu Ming ’s treatment of CVI is mainly based on raising Yang and dehumidification,and is based on invigorating qi and raising Yang,promoting Yang with fengyao,inducing diuresis with bland drug,as well as promoting blood circulation and removing stasis.While strengthening qi and eliminate pathogenic,attention should be paid to the lifting cycle of the body,and ascending and descending are used simultaneously,mainly ascending in combination with the characteristics of CVI in lower limbs.2.The core prescription for CVI treatment involves luteolin,quercetin,β-sitosterol,stigmasterol and other main active ingredients;TNF,IL-6,IL-1β,VEGFA,PTGS2 and other core targets;Cancer pathway,PI3K-Akt signaling pathway,TNF signaling pathway,etc.Traditional Chinese medicine has the characteristics of multi-component,multi-target and multi-pathway.This study is expected to provide support for further research. |
