Objective: Through network pharmacology,the research methods of drug,gene and protein interaction were integrated data and the complex network between drug-target-disease was constructed to analyze the mechanism of Huazhuo Jiedu Shugan Decoction in treating epilepsy.Methods: This study used TCMSP and BATMAN-TCM database collection of Huazhuo Jiedu Shugan Decoction’s 6 flavour of herbal active ingredients and targets,with the STRING database in the traditional Chinese medicine database name into the target protein gene symbal,epileptic disease through input keywords "epilepsy" in Dis Ge NET database collection targets for epilepsy disease,using Jvenn tools take intersection of the Huazhuo Jiedu Shugan Decoction and epileptic disease,get Huazhuo Jiedu Shugan Decoction potential targets for treatment of epilepsy,the potential targets for import STRING database to predict protein-protein interactions,using Cytoscape constructing PPI network diagram and the network topology analysis,screening key targets.According to the various herbal active ingredients,and the relationship between the target to build herbal compound-target network,for explicit Huazhuo Jiedu Shugan Decoction targets of biological function and the degree of enrichment of signaling pathways,use a database of DAVID to GO and KEGG enrichment analysis,through the Cytoscape software will target and associated visualization and topology analysis of signaling pathways,obtain the core targets and its role in signaling pathways,in order to analyze Huazhuo Jiedu Shugan Decoction mechanism of action of epileptic curative effect.Result: In this study for 6 flavour herbal composition Huazhuo Jiedu Shugan Decoction party on a total of 70 eligible for screening of effective components,baicalin,stigmasterol,rhizoma kaempferiae phenol,quercetin and sitosterol,anethole,cinnamic acid methyl ester,the methoxyl cinnamic acid,beta sitosterol,han within such as statins.Corresponding effective component of drug targets,394,1215 epileptic disease targets,the 135 potential targets of Huazhuo Jiedu Shugan Decoction treat epilepsy including MAPK1,RELA,TNF,IL6 and AKT1,IL1 B,TGFB1,FOS,MAPK8,IFNG,JUN,CASP3,PRKCA,CASP8,PRKCB,BCL2 and IL10,CCL2,IL1 A a total of 19 core targets.Involving biological features: response to hypoxia,nitric oxide biosynthesis of positive adjustment,aging and active control of gene expression,positive regulation of cell proliferation and response to lipopolysaccharide,combined the same protein,enzyme,transcription factor binding,nitric oxide synthase activity and plasma membrane adjustment and postsynaptic membrane,the cell surface,etc.And Huazhuo Jiedu Shugan Decoction side effect associated with epilepsy targets KEGG signaling pathways including the neural activity of ligand receptor interaction,cholinergic synapses,serotonin synaptic synapses related pathways,such as TNF,HIF-1signal pathway.Conclusion: Baicalin,stigmasterol,kaempferol,quercetin,sitosterol,anethole,methyl cinnamate,p-methoxycinnamic acid,β-sitosterol,isohanate and other key compounds in Huazhuo Jiedu Shugan Decoction are involved in apoptosis,anti-inflammatory mechanisms,anti-oxidative stress,and regulation of neurotransmitters.It is speculated that they may act on core targets such as MAPK1,TNF,IL6,AKT1,IL1 B,TGFB1,FOS,MAPK8,CASP3,and CASP8,regulates neuroactive ligandreceptor interactions,cholinergic synapses,serotonergic synapses,TNF signaling pathways,HIF-1 signaling pathways and other signaling pathways to exert anti-epileptic effects.
