| Objective:To explore the improving effect of Xiangqin Jiere Granule(XQ)on obese mice induced by different factors and its possible mechanism,To explore the effect of Xiangqin Jiere Granule(XQ)on lipid metabolism and its possible mechanism in obese mice induced by different factors,the obese mice model with monosodium glutamate(MSG)and high fat diet(HFD)were established.Methods:Part one: The obese mice model of monosodium glutamate(MSG)was established by subcutaneous injection of MSG solution(3mg/kg)on the 2nd to 8th day after birth in newborn Balb/c mice.The control group was subcutaneously injected with the same amount of saline,and after 21 days of age,the male mice were selected and fed in baskets.At the age of 12 weeks,the mice were randomly divided into three groups: Model of obese mice induced by MSG group(MSG group),Xiangqin jiere granule low dose group(XQ4.5 group,4.5g/kg),Xiangqin jiere granule high dose group(XQ22.5 group,22.5g/kg).Control group and MSG group were given the same amount of saline,XQ4.5 group and XQ22.5 group were given corresponding dose of Xiangqin jiere granule,continuous intragastric administration for 5 weeks,and the weekly body weight,abdominal circumference and food intake of mice were monitored.After fasting for 12 hours,the fasting weight,abdominal circumference,body length and Lee index were measured,visceral fat weight,liver tissue weight and thymus tissue weight were accurately weighed,and visceral fat percentage,liver index and thymus index were calculated.The levels of triglyceride(TG),total cholesterol(TC)and low density lipoprotein cholesterol(LDL-C)in serum and triglyceride(TG),total cholesterol(TC),low density lipoprotein cholesterol(LDL-C)and high density lipoprotein cholesterol(HDL-C)in liver tissue of mice were detected by kit.Detection of gene expression of lipid metabolizing enzymes ACC1,FAS,DGAT1 and HTGL in mouse liver by RT-q PCR,the levels of tumor necrosis factor(TNF-α)and interleukin 6(IL-6)in serum were detected by ELISA kit,and TNF-α and IL-6 in liver tissue were detected by RT-q PCR.Part two: Kunming male mice aged 6-8 weeks were given a high-sugar,high-salt and high-fat diet,and the body weight and abdominal circumference of mice were monitored every week.After successful modeling,the rats were randomly divided into high-fat diet obesity model group(Model group),low-dose Xiangqin Jierre granule group(XQ4.5 group,4.5g/kg),medium-dose Xiangqin Jierre granule group(XQ13.5 group,13.5g/kg)and high-dose Xiangqin Jierre granule group(XQ22.5 group,22.5g/kg).Normal mice were divided into Control group(saline)and Control+XQ group(Xiangqin jiere granule,13.5g/kg).saline and corresponding dose of Xiangqin jiere granule were given by intragastric administration for 5 weeks,and the body weight,abdominal circumference and food intake were monitored every week.After fasting for 12 hours,the fasting weight,abdominal circumference,body length and Lee index were measured,fasting blood glucose and glucose tolerance were measured,visceral fat weight,liver tissue weight and thymus tissue weight were accurately weighed,and visceral fat percentage,liver index and thymus index were calculated.The levels of TG、TC、LDL-C and HDL-C in serum and liver were detected by kit,The gene expression of lipid metabolizing enzymes ACC1,FAS,DGAT1 and HTGL in liver were detected by RT-q PCR.The protein levels of AMPK,p AMPK and SREBP-1c in liver were detected by Western blot.The gene expression of appetite stimulating neuropeptide Y(NPY)and proopiomelanocortin(POMC)in hypothalamus by RT-q PCR.The levels of tumor necrosis factor α(TNF-α)and interleukin 6(IL-6)in serum were detected by ELISA kit,and and the gene expression levels of TNF-α and IL-6 in liver tissue were detected by RT-q PCR.Results:Part one:The body weight,weight gain,abdominal circumference and Lee index of MSG obese mice decreased significantly(P<0.01,P<0.05,P<0.01),visceral fat and visceral fat percentage decreased significantly(P<0.01,P<0.01).The levels of serum TG,TC and LDL-C decreased(P<0.01),the level of TG in liver decreased significantly(P<0.01),the level of TC and LDL-C did not change significantly,the level of HDL-C increased,and the levels of ACC1,FAS and DGAT1 m RNA in liver decreased significantly(P<0.01),and the level of HTGLm RNA increased significantly(P<0.01).The level of IL-6 in serum and the level of IL-6m RNA in liver tissue decreased,but the level of TNF-α in serum and hepatic TNF-α m RNA increased significantly(P<0.01).Part two: After the obese mice with high-fat diet were treated with Xiangqin jiere granule for 5 weeks,the body weight and body weight growth of HFD obese mice decreased significantly(P<0.01),the abdominal circumference and Lee index decreased significantly(P<0.05),and the spleen index decreased significantly(P<0.05),but the thymus index and liver index did not change significantly.Visceral fat weight and percentage of visceral fat decreased significantly(P<0.01),fasting blood glucose and glucose tolerance decreased significantly(P<0.05),serum levels of TG,TC and LDL-C decreased significantly(P<0.01,P<0.01),and HDL-C did not change significantly.The levels of TG and TC in liver decreased significantly(P<0.01),the level of LDL-C increased significantly(P<0.01),the level of HDL-C decreased but there was no statistical difference,the gene expression levels of lipid metabolic enzymes ACC1,FAS and DGAT1 decreased significantly(P<0.01),and the gene expression of lipase HTGL enzyme increased significantly(P<0.01).The level of p AMPK protein and p AMPK/AMPK increased significantly(P<0.05),the level of SREBP1 c decreased,but there was no significant difference(P>0.05),and the levels of NPY,POMCm RNA and NPY/POMC decreased significantly(P<0.05,P<0.05,P<0.01,P<0.01).The level of serum TNF-α increased significantly,the level of liver TNF-α m RNA increased,but there was no statistical difference,the level of serum factor IL-6decreased,and the level of liver IL-6m RNA decreased significantly(P<0.01).The results of pathological sections of liver tissue showed that the liver cells of HFD obese mice had severe steatosis with a large number of spherical fat droplets,while the liver cell steatosis of HFD obese mice was significantly improved and the number of spherical lipid droplets decreased significantly after being treated with Xiangqin antipyretic granule.Conclusion:The results show that Xiangqin jiere granule has a good regulatory effect on lipid metabolism in MSG obese mice and HFD obese mice,and can effectively reduce body fat and inhibit body weight growth in obese mice.At the same time,Xiangqin jiere granule can reduce the food intake of obese mice with high-fat diet,inhibit the appetite of mice,and improve the chronic low-grade inflammation associated with obesity.At the molecular level,Xiangqin jiere granule can activate the AMPK/SREBP1c/ACC1 signal pathway of obese mice,regulate the level of fat metabolic enzymes,thus regulate lipid synthesis and metabolism,and reduce fat accumulation in the body. |
