Osteoarthritis(OA)is a kind of joint disease that occurs frequently in middle-aged and elderly people.It is joint pain and is accompanied by a reduction in the range of motion of the joint.If it is not intervened in time,it will develop to the end stage,the joint space of the patient will be narrowed,and in severe cases,deformity may occur,which will cause great pain to the patient.Schisandra chinensis(SF)has been shown to have therapeutic effects on osteoarthritis.Schisandra chinensis is a commonly used Chinese medicinal material.The main components are lignans,polysaccharides,organic acids and various lipids.The main active components are lignans.However,there is no clear study to confirm the pharmacological mechanism of Schisandra chinensis in the treatment of osteoarthritis.Based on the existing research on osteoarthritis and Schisandra chinensis,this paper uses bioinformatics,network pharmacology and molecular docking technology to predict the six most likely curative lignan compounds in Schisandra chinensis,and analyze the drug targets by constructing a PPI network and KOA targets,and identified 15 central genes(HUB genes)in the lignan-osteoarthritis target protein interaction network.With the help of GO analysis and KEGG pathway analysis,the relationship between the treatment of osteoarthritis and the lignans in Schisandra chinensis was analyzed,and the related genes and pathways were found.Finally,the anti-OA effects and underlying mechanisms of lignans identified in the network pharmacology analysis were validated by molecular docking techniques.The biological processes of genes and proteins targeted by lignans in the treatment of osteoarthritis include immune responses,inflammatory responses,cell signaling and phospholipid metabolism.Network pharmacology and molecular docking results showed that lignans inhibited the expression of EGFR and MAPK14 genes by inhibiting the expression and activity of SRC genes,further reduced the expression of MMP13,and then promoted the expression of type II collagen,thereby exerting anti-OA.effect.The purpose of this study is to provide a theoretical basis for further research on the potential mechanism of lignans in Schisandra chinensis in the treatment of osteoarthritis by computer using network pharmacology and molecular docking technology,and to provide new ideas for the treatment of early osteoarthritis. |
