| Dictamnus dasycapus Turcz is a traditional medicine.At present,Dictamnus dasycapus Turcz decoction pieces are listed in Processing Standard of Chinese Medicinal in Jilin Province(1986),Processing Standard of Chinese Medicinal in China(1988)and Chinese Pharmacopoeia(2020),because there is no specific processing technology parameters,which has many inconveniences in production and quality control.Therefore,in this paper,based on the design of response surface combined with ahp weighted score,multiple verification and optimization of the processing technology were carried out by constructing the analysis method of infrared fingerprint assisted by multivariate chemometrics,and the quality standard was studied.The main conclusions are as follows:(1)Extraction technology: On the basis of single factor,combined with analytic hierarchy process comprehensive scoring method for response surface experiment design,results showed that the optimal extraction process: extraction temperature was80°C,solid-liquid ratio was 1:15(g/m L),extraction time was 1.5 h,ethanol concentration was 70 %,and extraction times were 1.This condition is the method of pretreatment for subsequent sample content determination.(2)Processing technology: The optimal processing technology were: add water ratio 1:1,stuffy and moist time 150 min,cutting specification 3 mm,drying temperature60°C,drying time 60 min.(3)Multivariate chemometrics method: Fourier transform infrared spectroscopy showed that the main absorption peaks of S1-S18 samples were basically the same with little difference.Second derivative infrared spectroscopy showed that the contents of S9 and S18 were significantly lower than those of other samples.The contents of S9,S18 and S11 in S1,S3,S4,S5,S6,S11 and S15 were higher,and the contents of ashenone in S1,S3,S4,S5,S6,S14,S16 and S17 were higher.The contents of flavone were higher in S1,S2,S3,S4,S5,S6,S10,S12 and S17.Dual-index sequence analysis showed that compared with other peaks,the common peak rates of samples S9 and S18 were lower and the variation peak rates were higher,and the common peak rates and variation peak rates were more prominent in 18 batches of samples.The results of similarity analysis showed that the correlation coefficients of samples S9-S18 with other samples are low around 0.914,and the similarity of S1-S5 groups is higher than0.994,indicating that all samples are similar.Cluster analysis showed that when the Euclidean distance was 10,each sample was divided into three categories,including S6,S14,S5,S1,S2,S8,S3,S15,S4,S13,S16,S11,S9,S10,S7,S12,S17,and S18 alone.The results showed that S1,S2,S3,S4 and S5 were the optimal processing conditions,the 5 groups are the optimal processes in the same processing conditions of response surface.(4)Quality standard: The character,microscopic and thin layer chromatography results showed that Dictamnus dasycapus Turcz medicinal materials and prepared slices all indexes were in line with pharmacopoeia standards.The water content of Dictamnus dasycapus Turcz medicinal materials and prepared slices was 6.67% and 7.60%,the mean total ash content of medicinal materials and prepared slices was 6.55% and 5.72%,the mean acid insoluble ash content was 0.74% and 0.370%,respectively.The contents of dictamnine,fraxinellone and obacunone were 0.541% and 0.643%,0.313% and0.405%,and 0.172% and 0.253%,respectively.Results The contents of three active components in processed decoction pieces were higher than those in raw,and all indexes were in line with pharmacopoeia standards,indicating that was proved that this cutting process was feasible.In this paper,the processing technology and quality standard of Dictamnus dasycapus Turcz were studied,as well as the condition of cutting technology was studied.The processing cost was reduced,the loss of active components was reduced,which was beneficial to the preparation and storage,and some reference basis were provided for the study of Dictamnus dasycapus Turcz and other medicinal materials. |
